Various antioxidative characteristics of total methanolic extract from Gardenia jasminoides were identified for the nitrite scavenging ability, ABTS (2,2’-azinobis (3-ethylbenzothizoline-6-sulfonic acid)) radical cation inhibition, SOD(superoxide dismutase) -like activity and elongation effect of lipid peroxidation using Rancimat. After successive partitioning with n-hexane, chloroform(CHCl3), n-butanol(n-BuOH) and water(H2O), each fraction showed the dose-dependent activity. n-BuOH fraction and H2O fraction showed the potent nitrite scavenging ability of IC50, 183 ppm and 194 ppm, respectively. As for the ABTS radical cation inhibition, the CHCl3 fraction was most potent and its IC50 was 159 ppm. SOD-like activity was slightly mild in all of the fractions and showed the maximum activity of 93.83% in 2000 ppm of H2O fraction. n-BuOH fraction showed the activity of 50.14% in the same concentration. The elongation effect of lipid peroxidation also increased dose-dependently and the antioxidative index of total methanolic extract was 2.93 in 1000 ppm, more effective than 1.66 of butylated hydroxy anisol in the same concentration. The active fraction was subjected to silica gel column chromatography and the compoundⅠand Ⅱ were isolated and identified as geniposide and crocin, respectively, by 1H-NMR spectral data. Geniposide and crocin showed the different nitrite scavenging ability, ABTS radical cation inhibition and SOD-like activity. The IC50 value of nitrite scavenging ability of geniposide was 940 ppm and crocin, 77 ppm. The IC50 value of the geniposide and crocin in ABTS radical cation inhibition was 684 ppm and 549 ppm, respectively.
