Glycoprotein of Cudrania tricuspidata Bureau (CTB) was isolated from CTB fruits which have been used to heal various disorders of the injury and lung as an herbal agent in Korea since long time ago. The CTB glycoprotein was identified to have a molecular weight of 75 kDa and consists of carbohydrate (72.5%) and protein moiety (27.5%). To know inhibitory ability of the CTB glycoprotein for inflammation mediated by reactive oxygen radicals, firstly anti-oxidative activity [1,1-Diphenyl-2-picrylhydrazyl (DPPH), superoxide anion, and hydroxyl radicals] of CTB glycoprotein was examined in cell free system, and then evaluated changes of inflammation-related signals [intracellular reactive oxygen species (iROS), nitric oxide (NO), nuclear factor-kappa B (NF-kB), cyclooxygenase (COX)-2, and inductible nitric oxide synthase (iNOS)], and pro-inflammatory cytokines [interleukin (IL)-1b], interleukin (IL)-6, and tumor necrosis factor (TNF)-a] in the lipopolysaccharide (LPS) (1 ug/ml)-treated RAW 264.7 cells. The results in this study showed that CTB glycoprotein (100 ug/ml) has a strong scavenging activity against DPPH, superoxide anion, and hydroxyl radicals without any pro-oxidant activity in vitro. Expressions of iROS, NO, NF-kB, COX-2, iNOS, and pro-inflammatory cytokines (IL-1b], IL-6, and TNF-a) were inhibited by treatment with CTB glycoprotein (50ug/ml) in the presence of LPS (1ug/ml). Data obtained from these experiments indicate that the CTB glycoprotein suppresses expression of inflammation-mediated factors (COX-2, iNOS, IL-1b], IL-6, and TNF-a) through regulation of NF-kB. Thus, this study provides evidence that CTB glycoprotein could be to use as an herbal agents for the treatment of inflammatory diseases.
