Phytochemical constituents were isolated from the Aerial Parts and Roots of Taraxacum coreanum (Asteraceae) through open column chromatography and prep-HPLC. And we also employed a standardized several Taraxacum species extracts to determine contents by HPLC. The dried and finely powdered aerial part of T. coreanum which was extracted with methanol under reflux at 65 ~ 75℃ and solvent was evaporated in vacuo to give brown residue. The residue was suspended in H2O and partitioned with n-hexane, CHCl3, EtOAc, and n-BuOH successively. A portion of the each fraction subjected to column chromatography over silica gel and eluted with gradients to afford sub fractions. Recrystallization and prep-HPLC of sub-fractions was filtered, and compounds were yielded. Their structures were identified as β-sitosterol, daucosterol, taraxasteryl acetate, chrysoeriol, diosmetin, luteolin, luteolin-7-O-glucoside, esculetin and 5-hydroxypyrrolidin-2-one by interpretation of spectroscopic analysis including 1H-NMR and 13C-NMR and MS. Among them, diosmetin and 5-hydroxypyrrolidin-2-one were isolated for the first time from the plant. Luteolin and luteolin-7-O-glucoside were the functional compounds that suppressed oxidative stress. Taraxacum species contains a relatively rich source of luteolin and luteolin-7-O-glucoside. Since we could not find previously described as constituents of T. corenum, we also employed a standardized several Taraxacum species extracts to determine its contents by HPLC. Luteolin and luteolin-7-O-glucoside standards were isolated and identified from previous experiment. A Nucleosil 100-5 C18 column was used for the determination of luteolin and luteolin-7-O-glucoside. The mobile phase was a mixture of ACN and water – 0.1% formic acid (99.9 : 0.1, v/v). The gradient elution program was 10 to 23% ACN within 60 min at the flowrate of 1.0 ml/min. The UV spectra were recorded 350 nm for the identification. Luteolin and luteolin-7-O-glucoside are high contents in T. officinale flowers (5.817 ± 0.030 mg∙g-1) and T. coreanum leaves (6.205 ± 0.087 mg∙g-1), respectively. Among the several different parts of Taraxacum species, the highest contents of luteolin were measured in flowers and luteolin-7-O-glucoside were measured in leaves. Total contents of luteolin and luteolin-7-O-glucoside in T. coreanum is higher ca. 1.3 ~ 2 times than those of T. ohwianum and T. officinale.
