Degree of hydrolysis was determined in three ways; 1) soluble protein in supernatant using lowry assay, 2) protein in precipitates using semimicro kjedahl, 3) gravimetric method using weight difference before and after enzymatic hydrolysis.
The rice wine meal (RWM) was enzymatically hydrolyzed using eight commercial proteases for 4hr at optimum pH and temperature. Three proteases, Protamex, Alcalase and Protease N, are found to be the most effective for enzymatic hydrolysis of RWM. Although, there was some gab among those three methodologies, they showed similar pattern. When three enzymes were applied to RWM at one time, no significant difference was observed compared to single protease application in lowry, kjedahl and gravimetric methods (P<0.05) indicating no synergistic effect of three kinds of proteases.
