PURPOSE: This study evaluated the use of a CD66c-specific sandwich enzyme-linked immunosorbent assay (ELISA) as a tumor marker to differentiate lung adenocarcinoma from other types of lung cancer. MATERIALS AND METHODS: The cross-reactivity of the anti-CD66c monoclonal antibodies (mAbs) 8F5 and AP11 with various carcinoembryonic antigen-related cell adhesion molecule antigens was investigated. CD66c in lung cancer cell lines and formalin-fixed, paraffin-embedded lung cancer tissues was quantified using a CD66c-specific sandwich ELISA. The antigen concentrations were determined using a bicinchoninic acid protein assay kit. RESULTS: Two mAbs (8F5 and AP11) against CD66c were used for the sandwich ELISA. The 8F5 mAb cross-reacted with the CD66c and CD66e antigens, whereas the AP11 mAb reacted only with the CD66c antigen. Sandwich ELISA using both 8F5 and AP11 mAbs gave a positive response for the A549 lung adenocarcinoma cell line, but yielded no response for other lung cancer cell lines or normal lung cells. The CD66c-specific sandwich ELISA was positive for 14/17 formalin-fixed, paraffin-embedded lung adenocarcinoma tissue specimens, but not for other lung cancer tissues, including squamous cell carcinoma and small cell lung cancer. With formalin-fixed, paraffin-embedded tissue, the CD66c-specific sandwich ELISA showed 82% sensitivity and 92% specificity for lung adenocarcinoma (p < 0.003).
