Meat quality is affected by many factors such as genetic effects of animal, production and environmental conditions. Specially, fatness is one of the most crucial parameters determining meat quality. Quantitative trait loci (QTL) related to fatness and meat quality traits, such as intramuscular fat content (IMF) and backfat thickness (BFT) have been identified on pig chromosome 6 (SSC6). Moreover, LEPR and FABP were considered as potential candidate genes for controlling these QTL. Recently, Growth arrest and DNA damage inducible protein 45 alpha gene (GADD45A) was reported as a powerful candidate gene for QTL of BFT and IMF on SSC6. Thus this study was conducted to identify molecular markers associated with meat traits. Porcine GADD45A gene including 2Kb of 5’ regulatory region was amplified and sequenced to detect polymorphisms. Initially, a total of 8 SNP sites were found at g.-205T>C, g.-930T>G, g.-1622A>G, g.196A>G, g.392C>A, g.955T>C, g.2580delT and g.3247A>T in the porcine GADD45A. Subsequently association study was carried out on a total of 694 Berkshire pigs. Among 8 SNPs, 4 SNPs showed significant associations with multiple meat quality traits in the Berkshire population. In g.196 A>G SNP, animals with the AA genotype had significantly higher least square mean values of driploss, cookingloss and shear force (SF) than animals with the GG genotype (P = 0.0001, 0.0017 and 0.0295, respectively). Conversely, pigs with the GG genotype showed significantly higher least square mean values of CIE_lightness (MC_L) than pigs with the AA genotype (P = 0.0043). In g.329C>A, animals with the AA genotype had significantly higher least square mean values of water holding capacity (WHC), driploss, cookingloss, CIE_yellowness(MC_b) and shear force (SF) than animals with the CC genotype (P = 0.0064, 0.0001, 0.0019, 00240 and 0.0383, respectively). Conversely, pigs with the CC genotype showed significantly greater least square mean values of CIE_lightness (MC_L) than pigs with the AA genotype (P = 0.0022). In g.955T>C, animals with the CC genotype had significantly higher least square mean values of driploss, cookingloss, MC_b and shear force (SF) than animals with the TT genotype (P = 0.001, 0.0022, 0.0257and 0.0351, respectively).
