Ever since the discovery that nesfatin-1/NUCB2 is involved in the control of the appetite and energy metabolism in the hypothalamus, many reports have shown its expression in various tissues, including the adipose tissue and the tissues found in the digestive system and reproductive organs. However, no data currently exists on the expression of nesfatin-1/NUCB2 and its regulation mechanism in the reproductive organs. Therefore, the purpose of this study was to examine whether nesfatin-1/NUCB2 and its binding sites are present in the mouse reproductive organs, and to find out the regulatory pathway of its expression. In this study, 5-week old female ICR mice were used to assess the expression of nesfatin-1/NUCB2 in the ovary and uterus during the estrous cycle. The expression of NUCB2 mRNA was examined by RT-PCR and qRT-PCR. The nesfatin-1 protein expression was then detected by western blotting. Immunohistochemical staining was performed to confirm the expression of nesfatin-1 protein and its binding sites. The expression was also investigated in the collected primary oocytes, secondary oocytes, degraded oocytes and cultured ovarian granulosa cells. Furthermore, in order to examine if the expression of nesfatin-1/NUCB2 in the uterus is regulated by gonadotropin or sex steroid hormone, ovariectomized mice were treated with PMSG, 17β-estradiol or progesterone. As a result, the analysis of RT-PCR, qRT-PCR and western blotting demonstrated that a large amount of nesfatin-1/NUCB2 was expressed in the ovary and uterus, and showed the highest level of expression in the estrous stage. Immunohistochemical staining in the ovary showed that nesfatin-1 proteins were localized in the theca, interstitial cells, and granulosa cells, and its binding sites were displayed at the theca and interstitial cells near the tunica albuginea. Additionally, nesfatin-1/NUCB2 was also expressed in the primary, secondary oocytes and cultured granulosa cells. However, its binding sites were not detected in the all stages of oocytes. In the uterus, the expression of nesfatin-1 protein was detected at the epithelial cells surrounding the uterine glands and endometrium. The binding sites were also located in the epithelial cells surrounding the uterine glands and neutrophils of the endometrium. In the experiment to examine whether the expression of nesfatin-1/NUCB2 in the uterus was affected by PMSG, 17β-estradiol or progestrone, the expression levels of nesfatin-1/NUCB2 in the uterus was not changed after the treatment with PMSG in ovariectomized mice. On the other hand, nesfatin-1/NUCB2 expression in the uterus was dramatically increased after treatment with 17β-estradiol. The present study demonstrated for the first time that nesfatin-1/NUCB2 is expressed in the ovary and uterus and its expression in the uterus is regulated by 17β-estradiol. This suggests that the change in nesfatin-1/NUCB2, during the stage of the estrous cycle, would affect the gonadal function. Consequently, these results indicate that nesfatin-1/NUCB2 may play an important role in the reproductive organs as a local regulator.
