본 연구에서는, 단백질 가수분해 활성을 지닌 유산균을 이용하여 두유 발효물을 제조하고, 그 발효물의 주름개선 효과에 대하여 연구하고자 하였다. 이를 위하여 김치나 젓갈 등 각종 발효식품으로부터 단백질 가수분해 활성이 있는 총 108주의 분리된 유산균 중 일정 수준 이상의 protease 활성을 지니는 29주를 선정하였다. 그 중 두유 단백질 분해능이 가장 높은 균주 MK1을 본 연구의 발효균주로 최종 선정하였고, 생리학적 특성 및 16S rRNA 유전자 염기서열 등과 같은 분자유전학적 분석을 통해 Lactobacillus paracasei MK1로 동정 및 명명하였다. L. paracasei MK1을 이용하여 두유의 최적 발효산물을 제조하기 위하여 pH, 적정 산도, 생균수 및 단백질 분해 pattern에 따른 최적화 연구를 실시하였다. L. paracasei MK1의 종배양액을 멸균한 두유에 2%(v/v)접종하여 발효시켰을 경우, 발효의 최적온도는 30℃이었으며, 이 때의 적정산도와 pH는 발효 초기 0.15%와 pH 7.31에서 24시간 발효 후 0.98%과 pH 4.56으로 변화하였다. 생균수는 초기 6.94 log CFU/mL에서 발효 후 9.03 log CFU/mL로 증가하였다. 두유의 최적 희석배수는 50%로서 적정산도는 배양 24시간 후 0.08%에서 0.45%로 증가하였다. 최적 pH는 6.0이었으며 탄소원으로 2%(w/v) glucose를 첨가하였을 때 가장 우수한 발효 pattern을 나타내었다. 발효 시간에 따른 단백질의 분해 pattern을 sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)로 확인한 결과 최적 발효 시간은 30시간으로 확인되었다. 최적화된 상기 발효조건으로 L. paracasei MK1을 이용하여 두유 원액 또는 50% 두유 희석액을 발효한 후 한외여과를 실시하여 30 kDa 또는 5 kDa 미만의 분자량을 지니는 여액을 얻어 시료로 사용하였다. 또한 두유 발효 후 flavourzyme 또는 protamex로 분해한 뒤 한외여과를 통하여 30 kDa, 5 kDa 미만의 분자량을 지니는 여액을 시료로 사용하였다. 한편 두유를 flavourzyme 또는 protamex로 분해한 후 L. paracasei MK1로 발효시킨 발효액을 한외여과를 통하여 30 kDa, 5 kDa 미만의 분자량을 지니는 여액을 시료로 사용하였다. 상기 제조한 총 34개의 시료를 이용하여 피부세포에 대한 생리활성 평가를 시행하였다. DPPH에 의한 항산화능을 측정한 결과 모든 시료는 처리 농도가 증가할수록 항산화능도 증가하여 농도의존적임을 확인하였으나, 시료간의 큰 차이는 없었다. MTT assay를 이용하여 세포 독성을 측정한 결과 섬유아세포(fibroblast)에는 높은 시료 농도에서는 오히려 영양원으로 이용되어 성장이 증가하는 것을 확인하였으나, 각질형성세포인 HaCaT 세포에는 시료마다 그 정도의 차이는 있었으나 농도가 높을수록 세포의 생존율이 감소하는 것을 관찰하였다. 상기 실험결과를 50% 두유 희석액을 이용한 시료들에 대한 2차 효능평가를 실시하였다. 2차 효능평가는 16가지 시료를 대상으로 procollagen 양과 TNF-α 생성량을 측정하였다. Procollagen 생성량은 fibroblast에 UV를 조사한 후 시료를 처리하는 광노화 모델에서 실험하였을 때 ascorbic acid를 처리한 양성대조구(198%)와 50% 두유 희석액을 flavourzyme으로 처리하고 L. paracasei MK1으로 발효한 후 한외여과를 통하여 5 kDa 미만의 여액을 얻은 시료(50SFMKUF5)는 procollagen 생성량이 164%로 양성대조군과 유사한 생성량을 나타내었다. TNF-α의 생성량을 측정한 결과, 50% 두유 희석액을 flavourzyme 또는 protamex로 처리한 후 발효한 시료나 발효 후 flavourzyme 또는 protamex로 처리한 시료를 한외여과로 5 kDa 미만의 분획을 얻은 시료의 TNF-α의 생성량은 최소 23%로 양성 대조군인 dexamethasone의 TNF-α 생성량(66%)보다 낮게 나타나 5 kDa 미만의 여액 시료는 항염증 효과가 우수함을 확인하였다. 따라서 상기 procollagen 생성량과 TNF-α 생성량 결과에 의해 주름 개선 피부 미용 효과를 지니는 시료로서 50SFMKUF5를 최종 선정하였다. 한편 5 kDa 미만 분획물 시료 5가지의 일반 성분분석을 실시한 결과, 조단백질의 함량은 4.30 g/100 g에서 0.30 g/100 g으로, 조지방의 함량은 2.40 g/100 g에서 0.00 g/100 g으로, 고형분 함량은 약 10배가 감소하였다. 최종 선정 시료인 50SFMKUF5의 총 유리 아미노산의 함량은 67.3 mg/100 g, 필수 아미노산의 함량은 36.2 mg/100 g으로 가장 높은 것으로 측정되었다. LC/MS-MS를 이용한 peptide 분석에서는 major peak가 분자량 1453.74 kDa으로 밝혀졌고, 그 아미노산서열은 APEFLKEAFGVN으로 확인되었으나 주름 개선에 관련한 지표물질로 판단하기는 어렵다고 사료된다. 한외여과에 의해 5 kDa 미만의 분획을 얻은 시료들의 총 질소함량은 0.34-1.60 mg N/100 mL, TCA 용해 질소함량은 0.12-0.82 mg N/100 mL로 측정되었으며, 환원당량은 1.28-1.67 μmol/mL로 측정되었다. 이로써 선정된 유산균인 L. paracasei MK1이 두유 발효에 효과적임을 확인하였고, 유산균으로 인한 두유 발효물이 세포 수준에서 주름 개선에 효능이 있는 것으로 확인되었다.

This study was to prepare soymilk fermented with lactic acid bacteria which shows protease activity and to examine its anti-wrinkle effect on human skin. Total 108 strains of protease-producing lactic acid bacteria were isolated from various fermented foods such as kimchi and jeotgal, and among them, 29 strains which have high protease activity were selected. From 29 strains, strain MK1 was finally selected as a fermentation strain for this study due to its high ability to soymilk protein digestion. Strain MK1 was identified and named as Lactobacillus paracasei MK1 by physiological characteristics and by molecular genetics such as nucleotide sequence of 16S rRNA gene. When seed culture of L. paracasei MK1 was inoculated in sterilized soymilk at the final concentration of 2%(v/v) at 30℃ of optimum fermentation temperature, titratable acidity and pH were changed from 0.15 to 0.98% and from pH 7.31 to 4.56, respectively. The viable cell number increased from 6.94 to 9.03 log CFU/mL during fermentation. Optimum dilution rate of soymilk was 50% and, in that condition, titratable acidity was changed from 0.08 to 0.45%. The optimum condition for the fermentation of soymilk with L. paracasei MK1 was as follows; 30℃ of growth temperature, 30 h of growth time, pH 6.0 of initial pH of the growth medium. Soymilk was fermented with L. paracasei MK1 in optimum conditions and then fractionated with ultrafiltration membrane of which molecular weight cut off were 30 or 5 kDa. Soymilk was treated with flavourzyme or protamex preceded by fermentation with L. paracasei MK1. Soymilk was also fermented with L. paracasei MK1 preceded by reaction with flavourzyme or protamex. Anti-wrinkle and anti-inflammatory activities of test samples on fibroblast and keratinocyte were examined. Anti-oxidative activity of soymilk samples measured by DPPH method was higher than those of non-fermented soymilk. Fibroblast and keratinocyte were treated with test samples for cytotoxicity, and the treatment resulted in the increase in the growth of fibroblast and the decrease in the survival rate of keratinocyte. Cell based test was done with procollagen and TNF-α by treating 50% soymilk, and its protective effect against ultraviolet (UV)-induced skin damages was investigated. Treatment with 50SFMKUF5(50% soymilk→flavourzyme→fermented with L. paracasei MK1→UF 5 kDa) sample resulted in the increase of procollagen synthesis by 164% in human dermal fibroblasts and the decrease of a pro-inflammatory cytokine, TNF-α(23%), induced by UV irradiation in human keratinocytes. 50SFMKUF5 was finally selected to investigated the effect of anti-wrinkle on skin care. When 100% soymilk was fermented to make 50SFMKUF5, crude protein and crude lipid were changed from 4.30 to 0.30 g/100 g and from 2.40 to 0.00 g/100 g, respectively. Solid content of 50SFMKUF5 decreased ten times less than 100% soymilk. Total nitrogen and TCA soluble nitrogen contents of filtrates prepared by ultrafiltration with 5 kDa MWCO were ranged 0.34-1.60 and 0.12-0.82 mg N/100 mL, respectively. Reducing sugar content was measured to 1.28-1.67 μmol/mL. This is the first study to demonstrate the effectiveness of fermented soymilk to inhibit the UV-induced skin damages. Thus, it is suggested that fermented soymilk may be successfully employed as an “anti-inflammatory” and also “anti-aging” agent derived from a natural resource.
