본 연구에서는 사자발쑥(Artemisia princeps Pampanini, A. princeps Pamp.)의 항산화능과 성분 분석 및 타이로시네이즈 저해활성을 통한 기능성 화장품으로서의 이용 가능성을 평가하였다. 또한, 사자발쑥 추출물의 피부 전달시스템으로 에토좀을 제조하여 입자 크기, 포집효율, 안정성 및 피부 흡수 증진 능력을 평가하였다. 사자발쑥 추출물의 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거 활성(FSC50)은 에틸아세테이트 분획(12.27 μg/mL)에서 높은 활성을 확인하였다. 또한 luminol-의존성 화학발광법을 이용한 Fe3+-EDTA /H2O2 계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 총항산화능은 사자발쑥 추출물 에틸아세테이트 분획(OSC50 0.33 μg/mL)이 L-아스코르빅산(1.50 μg/mL) 보다 약 5배 더 큰 활성을 나타냈다. Rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 사자발쑥 추출물의 세포보호 효과를 측정하였을 때 아글리콘 분획에서 농도 의존적으로 보호효과를 나타내었다. 사자발쑥 추출물 에틸아세테이트 분획의 타이로시네이즈 저해 활성(IC50)은 29.20 μg/mL로 미백제로 알려진 알부틴(226.88 μg/mL)보다도 약 7배 정도 더 큰 활성을 나타냈다. 피부 상재균에 대한 항균활성 측정결과, 사자발쑥 추출물의 에틸아세테이트 분획에서 Gram (+) 세균인 Staphylococcus aureus, Bacillus. subtilis 및 Propionibacterium acnes에서 큰 항균활성을 나타냈다. 에틸아세테이트 분획을 TLC, HPLC, LC/ESI-MS/MS로 분석한 결과, 항산화 물질인 eupatilin과 jaceosidin이 있음을 확인하였다. 0.06 % 사자발쑥 에틸아세테이트 분획을 담지한 에토좀의 입자 크기는 286.8 nm, 포집효율은 51.96 %으로 나타났다. 0.06 % 사자발쑥 에틸아세테이트 분획을 담지한 에토좀은 제조 후, 3주 동안 단분산 형태를 유지하며, 사이즈의 변화가 거의 없었다. 피부투과 실험 결과, 에토좀은 일반 리포좀이나 에탄올 용액에서 보다도 더 큰 피부 투과능을 나타냈다. 이상의 결과로써 사자발쑥 추출물이 1O2혹은 다른 ROS를 소광시키거나 소거하고, ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 시사한다. 또한 피부 전달하기 위한 제형으로서 에토좀의 적합성과 항산화, 항노화 기능성 화장품 제형으로서의 응용 가능성을 검토하였다.

In the present study, the antioxidative and antibacterial activities of Artemisia princeps Pampanini (A. princeps Pamp.) extracts were investigated. Ethosome for enhanced transdermal delivery were prepared. Size, loading efficiency, stability, and cumulative permeated amounts of ethosomes were evaluated. The ethyl acetate fraction of A. princeps Pamp. showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity (FSC50 = 12.27µg/mL). Reactive oxygen species (ROS) scavenging activities (OSC50) of A. princeps Pamp. extracts on Fe3+-EDTA/H2O2 system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of extracts (OSC50 = 0.33 µg/mL) showed 5 times more excellent ROS scavenging activity than L-ascorbic acid (1.50 µg/mL) known as a water soluble antixoidant. The cellular protective effects of fractions of A. princeps Pamp. on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The aglycone fraction of extracts suppressed photohemolysis in a concentration dependent manner. The inhibitory effects of A. princeps Pamp. extracts on tyrosinase were investigated to assess their whitening efficacy. The ethyl acetate fraction showed 7 times higher tyrosinase inhibitory effect (IC50 = 29.20µg/mL) than albutin known as a whitening agent. Antibacterial activity of ethyl acetate fraction against various skin normal flora was measured. The results showed that the antibacterial activity of the fraction was the highest on the Staphylococcus aureus, Bacillus. subtilis, and Propionibacterium acnes. Antioxidant substances were isolated and purified from the ethyl acetate fraction. Eupatilin and jaceosidin were identified. The size of 0.06 % A. princeps Pamp. ethyl acetate fraction loaded ethosomes was 286.8 nm and the loading efficiency was 51.96 %. The ethosome loaded with 0.06 % A. princeps Pamp. ethyl acetate fraction was stable and maintained the constant particle size for 3 weeks after being prepared. The ethosome exhibited more enhanced skin permeability than general liposome and ethanol solution. These results indicate that the extract/fractions of A. princeps Pamp. can function as antioxidant and/or antibacterial agent in skin. I propose that ethosome has potential to be a transdermal delivery system for the anti-aging and antioxidant cosmetics.
