고구마(Ipomoea batatas L.)는 전 세계적으로 보리와 쌀 등의 곡류와 함께 주요 식량자원으로 이용되어 왔으며, 뿌리 뿐 만 아니라 지상부 역시 천연의 β-carotene과 안토시아닌, 무기물과 각종 비타민 그리고 양질의 식이섬유를 함유하고 있다. 아시아, 아프리카 등 지역에서는 잎과 잎자루를 채소로 이용하고 있으며 다량의 단백질을 함유하고 있어 단백질 사료로의 가능성이 제기되기도 하였지만 지상부에 대한 이용은 아직도 미미한 실정이다. 따라서 본 연구에서는 고구마 지상부의 다양한 효능을 이용한 식품 소재로서의 가능성을 조사하기 위하여 부위별로 이화학적 특성을 살펴보고 유용성분인 lutein, total polyphenol, chlorophyll을 효과적으로 추출하기 위하여 반응표면분석을 이용하여 추출조건을 최적화 하였으며 연구결과는 다음과 같다. 1. 고구마 잎과 잎자루의 품종별 부위별 이화학적 특성 고구마 잎과 잎자루의 조단백질과 조회분 함량은 품종별 부위별로 유의적인 차이를 보였으며 조단백질은 잎(15.57∼20.34%)이 잎자루(3.71∼5.97%)에 비하여 훨씬 높았으며, 연황미 잎에서 가장 높게 나타났다. 조회분 함량은 잎 (9.81∼14.91%)이 잎자루 (10.17∼15.39%)에 비하여 높았으며, 진홍미 잎에서 가장 많은 함량을 나타내었고, 무기성분은 주로 K과 Ca가 함유되여 있었으며, 그 다음으로 K, Mg, Al, Fe, Na, Mn, Zn 및 Cu이 미량으로 존재하였다. 고구마 잎과 잎자루의 lutein, β-carotene, chlorophyll 및 tannin 함량은 모두 품종별 그리고 부위별로 유의적인 차이를 나타내었다. Lutein 함량은 잎의 경우 190.1∼288.5 μg/g 범위로 헬씨미에서 가장 높게 나타났고 잎자루의 경우 18.8∼37.7 μg/g 범위로 건미에서 가장 높게 나타났다. β-carotene은 잎의 경우 352.1∼520.1 μg/g 범위로 헬씨미에서 가장 높게 나타났고 잎자루의 경우 4.8∼25.7 μg/g 범위로 신건미에서 가장 높게 나타났다. Total chlorophyll 함량은 잎의 경우 440.9∼712.2 mg/100g 범위로 헬씨미에서 가장 높게 나타났고 잎자루의 경우 36.8∼81.1 mg/100g 범위로 연황미에서 가장 높게 나타났다 . Tannin 함량은 잎의 경우 22.8∼44.6 mg/g 범위로 건풍미에서 가장 높게 나타났으며, 잎자루의 경우 6.8∼13.9 mg/g으로 신천미에서 가장 높게 나타났다. 항산화 성분과 항산화 활성을 측정한 결과 잎의 총 폴리페놀 함량과 플라보노이드 함량은 각각 46.26∼85.10 mg GAE/g 및 40.25∼68.37 mg CE/g 범위로 건풍미에서 가장 높았으며, 잎자루는 각각 14.60∼37.93 mg GAE/g 및 10.18∼22.48 mg CE/g로 신천미에서 가장 높았다. DPPH 라디칼 소거활성은 잎의 경우 건풍미와 증미에서 IC50 값이 0.109 mg/mL로 가장 높았으며, 잎자루는 신천미가 0.448mg/mL로 가장 높았다. 총 항산화력은 잎의 경우 건풍미가 58.75 mg AA eq/g으로 가장 높았고 잎자루는 대유미가 14.95 mg AA eq/g으로 가장 높았다. 2. 고구마 잎의 유용성분 추출 조건의 최적화 추출온도, 용질의 농도 및 용매량에 따른 lutein, total polyphenol 및 total chlorophyll 함량과 항산화활성에 대한 반응표면 분석한 결과와 최적 조건의 예측 결과는 다음과 같다. 3개의 종속변수 모두 추출온도와 용매 비 보다는 에탄올 농도에서의 영향을 크게 받는 것으로 나타났다. 루테인 함량은 3개의 요인변수의 영향을 모두 받는 것으로 나타났지만 에탄올 농도와 용매 비에서는 0.1%의 유의수준에서 영향을 크게 미치는 것으로 나타났다. 총 페놀성 화합물 함량은 에탄올 농도가 높을수록 증가하다가 70% 이상에서는 감소하는 것으로 나타났으며, 추출온도보다는 에탄올 농도에 더 큰 영향을 받는 것으로 나타났다. 클로로필 함량은 에탄올 농도가 높을수록, 용매량이 많을수록 함량이 증가하였으며 추출온도에는 큰 영향을 받지 않는 것으로 나타났다. 전자공여능은 추출온도보다는 에탄올 농도와 용매량에서 모두 0.1% 유의수준에서 큰 영향을 받는 것으로 나타났다. 총 항산화력은 용매비와 에탄올 농도에서는 모두 0.1% 유의수준에서 큰 영향을 받는 것으로 나타났으며 추출온도에서는 거의 영향을 받지 않는 것으로 확인되었다. 2-1. Lutein 함량의 최적 조건은 추출온도, 용매 비 및 에탄올 농도 각각 46.75℃, 25.78 mL/g 및 81.23%로 최대 381.88 μg/g으로 예측되었으며, 동일조건에서 실제 실험하여 얻은 값은 367.56 μg/g 이었다. 2-2. Total polyphenol 함량의 최적 조건은 추출온도, 용매 비 및 에탄올 농도 각각 53.90℃, 29.10 mL/g 및 67%로 최대 58.44 mg/g으로 예측되었으며, 동일조건에서 실제 실험하여 얻은 값은 55.21 mg/g 이었다. 2-3. Total chlorophyll 함량의 최적 조건은 추출온도, 용매 비 및 에탄올 농도 각각 69.90℃, 25.98 mL/g 및 93.10%로 최대 769.10 mg/100g으로 예측되었으며, 동일조건에서 실제 실험하여 얻은 값은 783.90 mg/100g 이었다. 2-4. 전자공여능의 최적 조건은 추출온도, 용매 비 및 에탄올 농도 각각 51.07℃, 33.01 mL/g 및 70.50%로 0.649 mg/mL으로 예측되었으며, 동일조건에서 실제 실험하여 얻은 값은 0.635 mg/mL 이었다. 2-5. 총 항산화력의 최적 조건은 추출온도, 용매 비 및 에탄올 농도 각각 44.52℃, 26.01 mL/g 및 64.42%로 최대 32.54 mg AA eq/g으로 예측되었으며, 동일조건에서 실제 실험하여 얻은 값은 33.62 mg AA eq/g 이었다. 이상의 결과를 종합해 보면 고구마 잎과 잎자루의 이화학적 특성을 살펴보고 유효성분의 추출 조건을 최적화함으로써 기능성 식품 소재 이용 시 기초자료로 활용될 수 있을 뿐만 아니라 폐기 처분되는 고구마 지상부의 이용을 확대시킴으로서 산업적으로 사용 시 재이용 가치를 확인할 수 있었고 경제적으로도 효과를 기대할 수 있을 것이다.

In order to use sweet potato leaves and stalks as functional food materials, we investigated the physicochemical characteristics of sweet potato leaves and stalks and optimization of functional components extraction condition. Fourteen cultivars were selected for this experiment. A significant difference was observed in the physicochemical characteristics of sweet potato leaves and stalks(p<0.05). For the present study, the mineral composition content in sweet potato leaves were significantly different among the cultivars(p<0.05). In particular, the major mineral was Ca and K. Crude protein content of leaves ranged between 15.57% in Gunpungmi and 20.34% in Yeonhwangmi and that of stalks between 3.71% in Yulmi and 5.97% in Shinchunmi. The lutein and β-carotene content were also high in leaves, and was ranged from 190.1∼288.5 μg/g and 352.1∼520.1 μg/g, respectively, while those of stalks were between 18.8∼37.7 μg/g and 4.8∼25.7 μg/g, respectively. The levels of lutein and β-carotene contents of leaves were high in Healthymi. Chlorophyll a predominated over chlorophyll b in partially colored leaves and stalks. Chlorophyll a, chlorophyll b and total chlorophyll were the highest (518.6 mg/100g, 193.6 mg/100 and 712.2 mg/100g, respectively) for Healthymi and the lowest for Gunmi (329.0 mg/100g, 111.5 mg/100 and 440.9 mg/100g, respectively) (p<0.05). Functional components from sweet potato leaves were extracted by ultrasound-assisted extraction technology. The experiments were carried out according to a three level, three variable central composite rotatable design. The best possible combinations of extraction variables were obtained for the maximum phenolic compounds, lutein and chlorophyll of sweet potato leaves extracts using response surface methodology. In the extraction conditions that were based on the central composite design with variations in the ethanol concentration, temperature and the ratio of the solvent to the sample. Among the rest, ethanol concentration instability has the most significant effect on all about of functional components (lutein, polyphenol and chlorophyll). The optimal conditions include 81.23% ethanol concentration, 46.75℃ temperature and 25.78 mL/g ratio of solvent for the maximum lutein content (381.88 μg/g) and 67.0% ethanol concentration, 53.90℃ temperature and 29.10 mL/g ratio of solvent for the maximum total polyphenols (58.44 mg GAE/g) and 93.10% ethanol concentration, 69.90℃ temperature and 25.98 mL/g ratio of solvent for the maximum total chlorophyll content (769.14 mg/100g) and 70.50% ethanol concentration, 51.60℃ temperature and 33.01 mL/g ratio of solvent for the maximum IC50 of EDA (0.649 mg/mL) and 64.42% ethanol concentration, 44.51℃ temperature and 26.01 mL/g ratio of solvent for the maximum AEAC (32.54 mg AA eq/g). Under these conditions, the experimental functional components of the sweet potato leaves extract, which is well matched with the predicted values. Consequently, the present study was carried out to contribute to the simulation and optimization of sweet potato leaves extraction.
