NO의 생성에 영향을 미치는 iNOS 단백질의 발현량을 측정한 결과 LPS 처리에 의해 활성화된 iNOS 단백질의 발현이 장미꽃 methanol 추출물 처리시 유의적으로 감소하였다. Luciferase assay 를 실행한 결과 LPS 로 자극한 세포와 비교하였을 때 염증과 관련된 NF-κB promoter activty 가 장미꽃 methanol 추출물 처리시 현저히 감소하는 경향을 나타내었고, 세포질의 IκBα의 인산화를 저해함으로써 전사 요소인 NF-κB p65, p50을 핵속으로 유리시키는 과정을 억제하였다. 이 결과로 장미꽃 methanol 추출물이 전사단계에서 저해 활성을 나타낸다는 것을 확인하였다. 본 연구 결과 장미꽃은 우수한 황산화 활성, 산화적 스트레스에 대한 보호효과 및 항염증 효과를 나타냄에 따라 만성질환 예방을 위한 기능성 식품의 원료로 활용될 수 있을 것으로 사료된다.

Rose is a woody perennial of the genus Rosa, within the family Rosaceae. They are known as edible flowers and have been consumed for many years as food components, such as cakes and beverages. They were also used in medicinal practices for remedy of various illnesses. Four different rose cultivars were chosen as the samples were determined. However, the antioxidant activity and anti-inflammatory effect are not yet fully understood. In the present study, we investigated the antioxidant activity of rose methanolic extracts (RMEs) and their protective effect involved in a tert-butyl hydroperoxide (TBHP)-induced oxidative injury in HepG2 cells and anti-inflammatory activity in LPS-stimulated RAW264.7 macrophage, respectively. First of all, we evaluated antioxidant compounds and activities of RMEs. Among the four different rose extracts, the methanolic extracts prepared from Oklahoma showed relatively higher antioxidant compounds and antioxidant activities compare with other rose cultivars. Then, the potential cytoprotective effects of RMEs against oxidative damages induced by tert-butyl hydroperoxide (TBHP) in HepG2 cells were evaluated. The level of ROS, cellular lipid peroxidation, glutathione (GSH), and antioxidant enzyme activities were measured as biomakers of cellular oxidative stress. HepG2 cells were treated with samples for 6 h at the same concentration (50 μg/mL) and then the cells were exposed to TBHP for 3 h. The incubation of HepG2 cells in the presence of TBHP led to approximately 65% decrease in cell viability. However, pretreatment of the cells with the samples at the concentration of 50 μg/mL increased cell viability. Cellular generation of ROS, formation of malondialdehyde (MDA), and depletion of GSH were reduced by RMEs. Moreover, TBHP treatment increased the level of glutathione peroxidase (GPx), catalase (CAT), and superoxide dismutase (SOD), however, pretreatment of 6 h with samples significantly decreased the level of them at the concentration of 50 μg/mL. In conclusion, a treatment of RMEs protects the cells against oxidative damage by modulating ROS production, GSH level, MDA generation, and antioxidant enzyme activities in HepG2 cells. These results indicate a positive response of the cell defend system to cope with a TBHP-induced oxidative stress. Finally, we investigated anti-inflammatory effect of RMEs on nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression in LPS-stimulated RAW264.7 macrophages. RMEs inhibited NO production and the reduction of NO production was accompanied by a reduction in iNOS protein expression. Furthermore, RMEs inhibited the LPS-stimulated DNA binding activity of nuclear factor-κB (NF-κB). These results indicated that RMEs inhibited NF-κB activation, which might block iNOS induction. These results suggested that RMEs had potent anti-inflammatory activity via suppression of NF-κB in RAW264.7 macrophages.
