C57BL/6N mice 6 주령으로 8주 동안 일반 식이, 콜레스테롤 식이, 콜레스테롤 식이 + 유산균, 콜레스테롤 식이 + 심바스타틴 등 4 그룹으로 나누어 실험을 진행하였다. 그 결과 43121그룹은 콜레스테롤 식이에 의해 혈중 콜레스테롤 증가 지표인 LDL cholesterol, total cholesterol 수준을 감소시켰고, H&E 및 Oil red O 염색을 통해 콜레스테롤 섭취로 인한 지질 축적을 감소시키는 것을 확인하였다. 또한 43121은 RT-PCR 검사 결과 콜레스테롤 분해와 관련된 인자인 CYP7A1의 m RNA 수준이 높아 지는 것을 보여 주었고, 콜레스테롤 합성과 관련된 인자인 HmgCoAr의 m RNA 수준은 낮아지는 경향을 나타내었다. 이상의 결과에서 43121균주는 콜레스테롤 섭취로 유도된 고 콜레스테롤혈증,피부의 지질침착에 대한 손상을 완화하는데 효과를 보였으며, 이를 통해 43121균주는 콜레스테롤에 의한 피부 손상을 예방하는데 큰 도움을 줄 수 있을 것으로 사료된다.

To find protective effects of skin inflammation, we investigated the protective effects of oxidative stress circumstances using lactic acid bacteria. In our study, 30 strains of lactic acid bacteria have screened for antioxidant and cholesterol lowering ability. Lactobacillus acidophilus ATCC 43121 has shown the highest scavenging activity and cholesterol assimilation and selected for further analysis. During in vivo study, the protective effects of Lactobacillus acidophilus ATCC 43121 on hypercholesterolemia states were assessed. The animals were divided into the following groups: (1) ND group, AIN76A diet without cholesterol, (2) HCD group, AIN76A with 1.25 % (w/w) cholesterol, (3) 43121 group, cholesterol diet with freeze-dried probiotics and (4) SV group, cholesterol diet with cholesterol lowering drugs. Total cholesterol, triglycerides, low density lipoprotein cholesterol, high density lipoprotein cholesterol in serum were measured. The structure changes of skin were investigated by H&E staining and Oil red-o staining. The mRNA expressions related with cholesterol metabolism and inflammation factor were investigated. Here we present evidence that 43121 could alleviate the lipid accumulation response in the skin that is triggered by excess dietary cholesterol. In conclusion, these results suggest that the indigenous L. acidophilus ATCC 43121 have the potential to be explored as a probiotics in the management of hypercholesterolemia and skin lipid regulation.
