In general, Peucedanum japonicum is used in traditional herbal medicine in Korea, Japan, Philippines, China, and Taiwan, and it has been reported to have a variety of biological activities. The induction of apoptosis in cancer cells has become a major focus of anti-cancer medicine development, and therefore, studied the apoptotic effects of the MeOH/CH2Cl2 extract of leafs of P. japonicum and its
components on HL-60 cells. Two coumarins, cis-3',4'-diisovalerylkhellactone (A) and (3'S,4'S)-3'-isovaleryl-4'-(2S-methylbutyryl)khellactone (B), were isolated by bioactivity-guided fractionation. Each experiment revealed that compound A and B increase sub-G1 phase, early-, late-apoptosis population, and disruption of mitochondria membrane potential. According to these data, compound A and B have cytotoxicity on the HL-60. These results suggest that compound A and B induce apoptosis on the HL-60 cell lines, and it has potential on the anticancer drug.
