Mulberry (Morus alba L.) is widely distributed in Asia and all parts of this tree including root barks, fruits, twigs and leaves are of great importance in traditional medicine. Mulberry leaves (ML) has been used in traditional medicine for the treatment of metabolic disorders. It has been reported to be rich in flavonoids that have different biological activities, including antioxidant and anti-diabetic activities. However, the antioxidant activity, blood circulation improvement and anti-diabetic capacity of ML extract (MLE) were not or few reported. Therefore, the purpose of this study was to investigate the antioxidant capacity of flavonoid isolated from MLE, the ameliorative activity of blood flow and the anti-diabetic effect of MLE. First section of the study was to determine of the content of flavonoids in MLE and to investigate the antioxidant capacity of its components astragalin (AG), kaempferol (KF) and quercetin (QC) in vitro. The results were demonstrating that ML efficiently protects human red blood cells (RBCs) against free radical-induced oxidative damage, and that QC and KF were the predominant antioxidants in ML. Moreover, AG had the greatest protective effect against 2,2'-Azobis (2-amidinopropane) dihydrochloride (AAPH)-induced oxidative hemolysis and glutathione (GSH) depletion in RBCs. Taken together, the results of the present study suggest that MLE as well as flavonoids from MLE have strong antioxidant activity against free radical-induced oxidative damage due to the prevention of GSH depletion, and may be useful in diminishing the damage to RBCs. Second section of the research explores if MLE and QC, KF, and resveratrol (RV) help blood circulation improvement in the rat model that has damaged arteries caused by ferric chloride (FeCl3). The result were showing MLE and its relevant substances such as QC, KF, and RV are effective in preventing blood vessels damage and thrombosis in the rats with carotid induced by FeCl3, significantly improving blood flow rate, blood vessel area and blood flow amount. Therefore, MLE and the relevant substances are effective in preventing and treating cardiovascular diseases. The last section of the study was designed to investigate the anti-diabetic effects of MLE on high fat diet (HFD) and streptozotocin (STZ)-induced type 2 diabetes in mice. MLE and QC significantly dropped increases in blood sugar and serum insulin in the diabetic mice fed with HFD and STZ. MLE and QC significantly dropped the total cholesterol (TC), low-density lipoprotein (LDL), and neutral fat and raised high-density lipoprotein (HDL) in the diabetic mice fed with HFD and STZ. The levels of oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT) and atherogenic index (AI) were decreased in the MLE and QC groups. The serum insulin and leptin concentrations were reduced in the MLE and QC groups. These results showed that MLE could decrease blood glucose level and lead to an amelioration in dyslipidemia states on HFD/STZ-induced type 2 diabetes mellitus (DM) in mice. These results suggest that MLE and its compounds were useful materials to improve or treat against metabolic diseases such as oxidative stress, thrombosis and type 2 diabetes.
