The establishment of standardized fermentation process of broccoli sprouts were conducted in a batch that the fermentation temperature is confirmed by experiments with the most ideal conditions for fermentation in 20℃ ~ 40℃.
The experiment was carried out in order to separate and purify the surface material and the active ingredient. The column was run using HP20 resin in order to separate and purify the surface material and the active ingredient about supernatant and sediment after the broccoli sprouts(45.17g) were extracted with ethanol. It was tested with dividing into four fractions from 100% water to 100% ethanol and was confirmed the antioxidant activity of each fraction. As a result, it was confirmed that the specific antioxidant activity shown in 50% ethanol fraction with all fermented broccoli and non-fermented broccoli.
DPPH radical scavenging activity measurement results of fermented fermentation materials and non-fermented broccoli sprout extract by type of saccharide, the high antioxidant effects could be confirmed in the case of total activity than separately the activity of the supernatant and the sediment among non-fermented extract of broccoli sprouts. As a result of measuring the antioxidant activity by type of fermented saccharide, it could be confirmed that there are some of the differences in antioxidant activity by type of saccharide being used in fermentation, and it could be affirmed that the broccoli sprout extract fermented with Glucose, maltose and sugar than other saccharides among them appeared somewhat high activity. It showed a difference depending on the date fermented by type of saccharide, it could be confirmed that a month is suitable in the experiment to establish the fermentation period. Once the fermentation is conducted in more than that, it could be confirmed that the fermentation process is being conducted such as acetic acid fermentation. Based on the results it could be seen that active substances may bring changes in activity by decomposition of a different material through the saccharide fermentation.
The fermentation pattern for the four saccharides was analyzed by HPLC analysis for 4 weeks fermentation products among fermented broccoli. In the analysis results it can be compared with being confirmed that the 8 minutes peak of glucose is relatively small whereas confirmed that the 8 minutes peak of Sugar, maltose, fructose is a similar amount. In the total polyphenol and flavonoid content measurement results it could be confirmed that the total flavonoid content and the total phenolic content is high in fractions from the 50% ethanol in the supernatant and sediment, and it could be seen that the fraction is high in the antioxidant activity.
Therefore, it is expected to be larger for utilization as a material of antioxidant in high value-added for fermented materials through the broccoli sprout in the industrial side.
