DPPH 활성 측정 결과 에탄올 추출물은 3.85%, 열수 추출물은 77.87%로 나타났다. 검은생강 추출물의 총 폴리페놀 함량은 에탄올 추출물에서 3.00 mg/g, 열수 추출물에서 4.60 mg/g이었으며, 총 플라보노이드 함량은 에탄올 추출물에서 71.06 mg/g, 열수 추출물에서 9.49 mg/g으로 나타났다. 5 mg/ml 농도의 검은생강 추출물을 0.1, 0.3, 0.5% 수준으로 첨가하여 호상 요구르트를 제조하였다. pH는 발효 4시간에 5.75∼6.04에서 6시간에 4.43∼4.74로 감소하였다. 적정산도는 발효 6시간에 0.90∼1.00이었다. pH와 적정산도는 처리구별 간의 유의적 차이가 없었다. 유산균수는 모든 처리구와 대조구에서 발효 시간이 지남에 따라 유의적으로 증가하였으며, 6시간에 그 수가 가장 높아져 8시간까지도 유지되었다. 점도는 열수 추출물을 첨가한 시료에서 대조구와 비교하여 모든 처리구에서 증가하였다. 에탄올 추출물을 첨가한 시료에서는 대조구와 비교하여 점도가 높게 나타났으나 0.5% 첨가구에서는 감소하였다(p<0.05). 대조구 요구르트는 Escherichia coli에서 항균활성을 보였으며 검은생강의 에탄올 추출물 0.3%, 0.5%를 첨가한 요구르트가 보다 높은 활성을 보였다(p<0.05). 검은생강을 첨가한 요구르트로부터 ABTS 라디칼 소거활성에서 에탄올 추출물 첨가구는 첨가량에 따라 유의적으로 증가하여 0.5% 첨가구에서 27.76%로 가장 높게 나타났다. 열수 추출물 첨가구는 0.3, 0.5%에서 대조구와 비교하여 유의적으로 증가하였으며, 0.3% 첨가구에서 43.94%로 가장 높게 나타났다. This study was carried out to investigate the quality characteristics and functional properties of yogurt prepared with ethanol and hot water extracts of black ginger. As a result of the analysis of general components of dried black ginger powder, moisture content was 11.86%, crude protein content was 7.43%, fat content was 5.21%, crude fiber content was 6.02% and ash content was 7.14%. Antimicrobial activities of black ginger extracts were not detected in Salmonella typhimurium (SK713), Staphylococcus aureus (SK741), Escherichia coli (SK772) and Listeria monocytogenes (SK728). In the ABTS radical scavenging activity of black ginger extract, the ethanol extract showed 77.22% whereas hot water extract was 93.06%. The DPPH activity of ethanol extract showed 3.85% whereas hot water extract was 77.87%. Total polyphenol content of black ginger extract was 3.00 mg/g in ethanol extract and 4.60 mg/g in hot water extract. Total flavonoid content was 71.06 mg/g in ethanol extract and 9.49 mg/g in hot water extract. Yogurt was prepared by adding 5 mg/ml of black ginger extract to the levels of 0.1, 0.3 and 0.5%. The pH of black ginger yogurt decreased from pH 5.75∼6.04 at 4 hours of fermentation to 4.43∼4.74 at 6 hours. The titratable acidity was 0.90∼1.00 at 6 hours of fermentation. There was no difference in pH and titratable acidity among the treatments. The number of lactic acid bacteria increased significantly with fermentation time in all treatments and control, and the number increased to 6 hours with keeping up to 8 hours. Viscosity increased in all treatments in hot water extract added samples compared to control. Sample with ethanol extract showed higher viscosity than control, but decreased with addition of 0.5%(p<0.05). Control yogurt showed antimicrobial activity against Escherichia coli. The stirred yoghurt containing 0.3% and 0.5% ethanol extracts of black ginger showed higher activity than the control(p<0.05). The ABTS radical scavenging activity of yogurt added with black ginger was significantly increased according to the addition amount of ethanol extract, and the highest value was 27.76% at 0.5%. The addition of hot water extract increased significantly at 0.3 and 0.5% compared with the control, and the highest at 43.94% at 0.3%. The DPPH activity was highest at 29.87% in 0.1% ethanol extract. The activity of hot water extracts was significantly higher than that of the control, and the highest activity was found in the 0.3% treatment group (85.22%).
