DEGs set별로 시간의 흐름에 따라서 같은 패턴으로 변화하는 유전자를 하나로 cluster로 묶어서 clustering 분석을 수행한 결과, 주로 cytoplasm에서 transport 기능을 하며, 분자적으로는 nucleotide binding에 관련된 유전자 중, 성숙을 촉진하는 유전자들의 up-regulation과 성숙을 방해하는 유전자들의 down-regulation을 ‘조추’에서 확인할 수 있었다. 과실 형질 관련 후보유전자의 발현량을 형질변화와 비교 분석한 결과, 탄수화물 대사에 관련된 UGPase-1과 AGPase-1, 탄닌 생합성 경로의 DkLAC1, 카로티노이드 생합성 경로의 CRTISO와 PDS, 에틸렌 생합성 경로의 ACS와 ACO는 ‘조추’의 조기 과실 성숙을 유도하는 유전자로 추정되었다. Transcription factor(TF)로는 ethylene responsive gene의 promoter에 결합하는 ERF3와 ACS의 promoter에 결합하는 MADS-box TF인 RIN, AGL, FUL, 그리고 climacteric type의 성숙을 조절하는 NAC가 ‘부유’, ‘조추’ 단감의 성숙 조절에 핵심적 역할을 하는 것으로 추정되었다. 이들 12개 유전인자간 긴밀한 cascade 형성과 상호 보완적 작용이 단감 성숙기를 결정짓는 것으로 추론되었다.

The present study was conducted to understand genetic mechanism that controls fruit growth and development by comparing major fruit traits and transcriptome at different developmental phases between a late-ripening cultivar ‘Fuyu’ and an early-ripening cultivar ‘Soshu’ in PCNA persimmon (Diospyros kaki Thunb.). Fruit diameter, weight, firmness, peel color, soluble/insoluble tannin concentration, and sugar concentration were observed at every 1 or 3 week until 18th weeks after full bloom (WAB) for ‘Soshu’ and 24th WAB for ‘Fuyu’ in 2016 and 2017. The results indicated that fruit ripening started earlier in ‘Soshu’ compared to ‘Fuyu’. Fruit diameter and weight showed a typical dual S growth curve in both cultivars, while in ‘Soshu’ the 2nd growth phase was shorter and the shift from the 1st to 3rd growth phase occurred faster compared to ’Fuyu’. Fruit firmness drastically decreased from 15 WAB in ‘Soshu’ and caused significant difference between cultivars. Fruit peel coloring progressed faster in ‘Soshu’ than ‘Fuyu’, and difference in color between cultivars appeared to be remarkable at 18 WAB. Soluble tannin concentration decreased rapidly at 12 WAB in both cultivars, while insoluble tannin concentration decreased slowly after sharp decrease at early growth stage. For sugar concentration in fruit, reduction point of sucrose started 9 weeks faster in ‘Soshu’. Transcriptome analysis was carried out for fruit flesh at every 3 weeks during 9-18 WAB. GO term analysis of the transcriptome indicated that catalytic or transport genes involved in controlling metabolism and growth in cells and cellular organelle were mainly expressed, while in KOG and KEGG analysis, the majority of expressed genes were related to signal transduction, nuclear structure, and starch and sucrose metabolism pathway. The DEGs analysis was conducted for DEGs set I in which fruit samples of every 3 weeks until 18 WAB were compared as treatment to the samples of 9 WAB as control, and for DEGs set II in which fruit samples of ‘Soshu’ were compared as treatment to ‘Fuyu’ as control at each WAB. For DEGs set I containing three comparison combinations, the number of up-regulated genes in treatment sample was highest at 18 WAB in ‘Fuyu’ and at 15 WAB in ‘Soshu’. For DEGs set II, the number of down-regulated genes and the number of up-regulated genes were higher in ‘Soshu’ compared to ‘Fuyu’ at 12 WAB and 15 WAB, respectively. In KEGG analysis, gene expression level for second metabolite biosynthesis and carbohydrate metabolism was detected at high level throughout the growth stages, and especially these genes in ‘Soshu’ were mostly down-regulated at 12 WAB, while they were mostly up-regulated at 15 WAB. In addition, cluster analysis of genes showing a similar expression pattern at each WAB indicated that genes for transportation in cytoplasm and nucleotide binding were up-regulated in ‘Soshu’, but down-regulated in ‘Fuyu’. In comparison of the expression pattern of fruit trait-related candidate gene to the phenotypic changes in traits, UGPase-1 and AGPase-1 for carbohydrate metabolism, DkLAC1, for tannin biosynthesis, CRTISO and PDS for carotenoids biosynthesis, ACS and ACO for ethylene biosynthesis were considered as the genes that may induce early fruit ripening in ‘Soshu’. In addition, as transcription factors(TFs), the ERF3 binding to the promoter of ethylene responsive gene, the MADS-box TFs RIN, AGL, and FUL, and the NAC controlling climacteric type fruit ripening were considered to play a key role for controlling fruit ripening in ‘Fuyu’ and ‘Soshu’. The formation of cascade and complementary action among these 12 genes may control the time for fruit ripening in PCNA persimmon.
