As a substitute for high-price ginseng, this study attempted to examine a possibility of the ferment extract of Panax ginseng sprout whose leaves and roots can be used together as a cosmetic ingredient with anti-oxidative and wrinkle-care effects. In terms of a test method, antioxidant activities were confirmed through total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity, ABTS radical scavenging activity and FRAP assay, using the Panax ginseng sprout fermented by Saccharomyces cerevisiae. In addition, the inhibition of ROS/NO generation and anti-inflammatory effects were measured in macrophage RAW 264.7 cells. To assess wrinkle-care effectiveness, the cytotoxicity of the extract was analyzed through MTT assay, and formation of collagen in the cells and inhibition of collagenase activity were tested. For the purpose of verifying its availability as a cosmetic ingredient, furthermore, stability and safety were tested (primary skin irritation test), and the results found the followings: The Panax ginseng sprout ferment extract revealed antioxidant activities in total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity, FRAP assay and ABTS radical scavenging activity. Moreover, anti-inflammatory effects were confirmed by measuring the inhibition of ROS/NO generation. At a cytotoxic test, no cytotoxicity was observed. In fact, the extract revealed anti-aging and anti-wrinkle effects of MMP-1. In fact, its safety has been confirmed through a primary skin irritation test without any irritation on the skin, and there were no changes in stability. Therefore, this study has confirmed a possibility of Panax ginseng sprout ferment extract as a cosmetic ingredient with antioxidant, anti-inflammatory and MMP-1-inhibitory effects.
