ADHD 동물 모델에 삼중 재흡수 억제제인 TRI5810 (1, 5, 10, 20mg/kg)을 급성, 아만성(7일) 및 만성(14일) 등의 다양한 기간 동안 처리하여 화합물이 최적의 효력을 나타나는 용량과 처리 기간을 확인하였다. 강제 수영 테스트(forced swimming test, FST)를 통해 화합물의 항우울제 효능을 평가한 결과, TRI5810의 최적 효력은 20 mg/kg을 2주 처리한 결과임을 확인할 수 있었다. OFT 시험 결과, MK-801 처리에 의해 증가된 과잉행동 (총 이동 거리, 운동 지속시간)이 TRI5810 처리에 의해 유의적으로 회복되었다. 과잉 행동 이외 ADHD의 나머지 두 가지 핵심 증상인 부주의나 충동성은 본 연구에서 사용한 동물 모델이나 행동 실험 평가 패러다임에서는 유효하지 않았기에 다른 ADHD 모사 동물모델이나 행동 평가법 등을 활용하여 TRI5810의 효력 평가에 대한 추가적인 연구를 진행하고자 한다. 본 연구 결과는 삼중 재흡수 억제제를 활용하여 ADHD 약물의 치료 효과 및 기존 치료제 대비 부작용이 나타나지 않는 등의 신규 치료제로서의 개발 가능성을 시사한다고 판단된다.

TRI5810 is a novel triple reuptake inhibitor and is used to verify its effectiveness as an ADHD therapeutics in this study. In this study, the behavioral effects of TRI5810 were investigated in MK-801-treated mice. Eight weeks old male ICR mice were administered with MK-801 (0.2 mg/kg), an NMDA receptor antagonist intraperitoneally to induce ADHD-like behaviors and observed for hyperactivity and impulsivity through an open field test (OFT) and a cliff avoidance test, respectively. The animals were treated with a triple reuptake inhibitor, TRI5810, at different doses (1, 5, 10, and 20 mg/kg) as acute, sub-chronic (7 days), and chronic (14 days) to find out which time confer preferential effect. The compound’s antidepressant efficacy was also investigated through a forced swimming test and the data showed that 20 mg/kg was an optimal dose. Overall, these results imply that TRI5810, one of the triple reuptake inhibitors, has therapeutics effects against, which should draw further interests as a potential new drug candidate
