제안 방법
HPLC 분석 시 전처리 과정 중 여과지 분쇄 효과를 확인하기 위해, 여과지 분쇄를 생략한 시료와 여과지를 분쇄한 시료의 분석 결과를 비교하여 Chl α와 carotenoids에 대한 분쇄효과를 살펴보았다.
Picoplankton 중 Synechococcus와 같은 작은 크기의 식물플랑크톤의 불완전한 추출에 대한 영향을 보기 위해 cyanophytes의 지표색소인 Zea 농도에 따른 RChl α 값을 살펴보았다.
각 지표색소의 peak 동정은 표준 색소와 Retention Time을 비교하여 결정하였다. 표준 색소는 chlorophyll α, chlorophyll b, peridinin, lutein, fucoxanthin, zeaxanthin, 19'-hexanoyloxyfucoxanthin, alloxanthin, violaxanthin, 19'-butanoyloxyfucoxanthin, diadinoxanthin, diatoxanthin (VKI, Denmark)을 구입하여 이용하였다.
분쇄 시 발생할 수 있는 시료의 손실을 보정하기 위해서 내부표준물질로써 canthaxanthin 50 μL를 첨가하였다(Wright et al., 1991).

동해 남서부 해역에서 2013년 9월 30일~10월 3일 동안 4일에 걸쳐 R/V 탐양호를 이용하여 2개 정선 14개 정점에서 해수 시료를 채취하여 그 중 40개의 시료를 이용해 광합성 색소분석을 실시하였다(Fig. 1). 탐양호에 장착된 CTD(SBE 911)의 Rosette sampling system에 Niskin 채수기를 부착하여 표준 수심(10 m, 20 m, 30 m, 50 m, 70 m, 100 m)에서 각각 해수 3 L를 채수하였다.
채수한 해수시료는 현장에서 GF/F 여과지(47 mm)로 여과하고 광분해를 막기 위해 알루미늄 호일로 싼 후 드라이아이스에 저장 하여 실험실로 운반하였다. 운반 후 시료 분석 전까지 초저온 냉동고(-80℃)에 보관하였다.

분석된 전 정점의 결과 중, 수직, 수평적인 농도 분석의 경향을 보기 위해서 D transect의 시료 분석 값을 이용하였다. D transect의 0 m(D1, D2, D4, D6, D7) 또는 10 m(D3, D5) 에서 모든 광합성색소에 대한 R값을 비교해본 결과, Chl α 의 경우 연안보다 외양에서 낮은 R값을 보였다(Fig.

표준 색소는 chlorophyll α, chlorophyll b, peridinin, lutein, fucoxanthin, zeaxanthin, 19'-hexanoyloxyfucoxanthin, alloxanthin, violaxanthin, 19'-butanoyloxyfucoxanthin, diadinoxanthin, diatoxanthin (VKI, Denmark)을 구입하여 이용하였다. 식물플랑크톤의 지표색소인 광합성색소의 명칭에 대한 약어(Table 1)는 SCOR의 규정에 따랐다(Jeffrey and Vesk, 1997).
혼합시료를 100 μL loop에 주입하여 HPLC를 이용해 광합성 색소를 정량 분석하였다. 표준색소의 정량 식은 Jeffrey에 의해 알려진 흡광계수 (Jeffrey et al., 1997)를 이용하여 Park and Park(1997)의 방법으로 계산하였다.
성능/효과
Chl α 농도에 따른 RChl α값에서, Chl α 농도가 낮을수록 큰 R 값을 예상했으나 실제로는 Chl α 농도가 높은 시료에서도 낮은 R 값으로 수렴하는 양상을 보였다(Fig. 2).
Chl α 뿐만 아니라 모든 광합성 색소들의 분석에 있어서 외양으로 갈수록 분쇄 전후의 농도차이가 커지는 것을 확인할 수 있었다.
Chl α의 농도에 따라 분쇄효과가 다르게 나타나는데, Chl α 농도가 높은 경우 분쇄효과가 크게 나타났으며, 낮은 농도의 Chl α 시료의 경우에는 분산 범위가 매우 큰 오차가 나타났다.

따라서 분쇄과정 생략 시, Chl α 농도는 2~76% 범위 내의 차이와 평균 45% 과소평가됨을 보였다.
