The MRM chromatograms of blank samples were compared with those of spiked samples with the analyte, and there were no interferences at the respective retention times of ketamine and dexmedetomidine. The detection and quantitation limits of the instrument were 0.6 ㎍/L and 2 ㎍/L, respectively. The limit of quantitation for the method was 10 ㎍/kg. The results of the recovery test on meat and bone meal, meat meal, and pet food showed ketamine in the range of 80.48-98.63 % with less than 5.00 % RSD, and dexmedetomidine in the range of 72.75-93.00 % with less than 4.83 % RSD. 제안 방법

시험법의 정확도 및 정밀도를 확인하기 위하여 무처리 시료에 혼합표준용액을 첨가하여 회수율 실험을 진행하였다. 회수율 실험은 정량한계, 정량한계 2배, 10배의 농도로 5반복 수행하였고 평균 회수율과 상대표준 편차(RSD, %, relative standard deviation)를 산출하였다. 기기상 검출 한계 및 정량한계는 이미 알고 있는 저농도 분석 대상 물질을 포함한 검체의 신호와 공시험 검체의 신호를 비교하여 구하였고, 검출한계는 3:1의 신호대 노이즈비가 산출되는 농도, 정량한계는 10:1의 신호대 노이즈비가 산출되는 농도로 선정하였다.
이론/모형
케타민과 덱스메데토미딘은 신속하고 정확한 분석을 위하여 LC-MS/MS로 분석하였으며, 전기분무 이온 화법(ESI)의 positive ion mode, multiple reaction moni- toring(MRM)mode 조건으로 분석되었다. 선구이온 (precursor ion)은 full scan mode에서 질량 스펙트럼을 확인하여 케타민과 덱스메데토미딘의 질량 값(exact mass, M)에 양성자(H+)가 결합된 [M+H] 형태인 m/z 238, 201의 선구이온을 확인하였다.
6. 유효성 확인
확립된 시험법은 식품의약품안전평가원의 가이드라인에 따라 선택성(Selectivity), 직선성(Linearity), 검출한계(Limit of Detection), 정량한계(Limit of Quantifi- cation), 정확도(Accuracy), 정밀도(Precision)를 확인하였다

사료 중 케타민과 덱스메데토미딘의 잔류조사 결과, 실제로 이 두 성분이 130 C 이상의 고온과 7기 압 이상의 고압에도 분해되지 않고 사료에까지 잔류하는 것이 확인되었다. Foertsch 등의 연구에서도 케타민을 다양한 열 환경에 6개월 간 보관한 결과, 온도에 대한 농도의 변화가 없음을 확인하였고, 반감기 역시 다른 화합물에 비해 6개월까지 느리게 분해됨을 확인하였다.
그리고 케타민과 같은 마취제는 처리된 후 다양한 조직에 널리 분포되지만 그 농도는 사용된 농도보다 낮고 독성 효과도 훨씬 적다는 사전연구 결과들이 있었다. 이번 연구에서도 이와 유사하게 케타민과 덱스메데토미딘이 육골분에서 낮은 농도로 잔류하고 있음을 확인하였다. 이후 조사한 국내 유통사료에서는 두 성분들의 잔류가 확인되지 않았지만 본 연구의 결과로, 화학적으로 안정한 구조를 갖고 있는 유해물질들에 대한 조사의 필요성이 대두되었다.
