< 천연물을 이용한 어린이용 육제품 자가 제조 기술개발>
본 연구는 천연물을 이용하여 어린이용 육제품 제조기술을 개발하였다. 주 수행내용은 소화흡수 증진 제품, 결핍 영양소 강화 제품, 간편 섭취용 제품을 개발하고, 품질분석과 저장성을 조사하였다. 그 결과는 아래와 같다.

< 천연물을 이용한 어린이용 육제품 자가 제조 기술개발>
본 연구는 천연물을 이용하여 어린이용 육제품 제조기술을 개발하였다. 주 수행내용은 소화흡수 증진 제품, 결핍 영양소 강화 제품, 간편 섭취용 제품을 개발하고, 품질분석과 저장성을 조사하였다. 그 결과는 아래와 같다.
1. 천연물을 이용한 소화흡수 증진 육제품 개발
본 연구의 목표는 어린이가 육제품 섭취 시 단백질 흡수율을 높이기 위해서 단백질 분해효소가 함유된 과일껍질을 육제품에 포함시키는 제조법을 개발하고 그 효과를 쥐 실험으로 구명하는 것이다. 먼저 제조법 설정을 위해 과일껍질 4종을 코팅하여 육제품에 첨가한 결과, 참다래와 파인애플 코팅물 첨가에 의해 프레스햄의 관능적 특성이 좋아졌으며, 조직적 특성 중 경도와 탄력성이 증가되는 효과가 있었다. 또한 30일 저장기간 동안 대조구와 비교해, 이화학적, 조직적, 관능적 특성에 큰 차이를 발견할 수 없었다. 실험쥐 모델실험에서는 코팅천연물 함유 프레스햄 급여가 단백질 소화율을 개선시키지는 않았지만 사료급여량을 증가시켜 같은 기간 동안 체중을 더 증가시키는 것으로 조사되었다. 따라서 육제품에 단백질 분해효소를 함유한 과일껍질을 함께 섭취시키는 것은 동물성 단백질 섭식 장애가 있는 아동에게는 소화를 도와줄 수 있을 것으로 생각된다.
2. 천연물을 이용한 어린이 결핍 영양소 강화 육제품 개발
본 연구에서는 어린이에게서 결핍될 수 있는 Ca, Fe, 비타민 A 함량이 높은 육제품을 제조 하기 위해 소, 돼지 간을 이용하여 제조하는 방법을 설정하고, 저장성을 평가하였다. 생 간 보다는 가열된 간을 사용해서 육제품을 제조해야 관능적인 특성이 좋아지는 것으로 나타났다. 가열된 간을 육제품 제조 시 첨가하는 지방의 50%를 대체하고, 품질과 저장성을 조사한 결과, 이화학적 특성과 일반성분, 조직감 특성, 관능적 특성, 미생물 변화에서는 대조구와 저장성에서는 큰 차이가 없었으나 소, 돼지 간 첨가 시 기호도가 떨어지는 경향을 보였다. 또한 지방산 패도와 단백질 변패도가 대조구보다 높아 추가적인 보완 연구가 필요한 것으로 생각된다.
3. 천연물을 이용한 어린이 간편 섭취용 육제품 개발
본 연구는 어린이가 간편하게 섭취할 수 있는 육제품 제조법을 개발하고자 하였다. 단백질 함량이 높은 돼지 뒷다리 부위와 소, 돼지 혀를 사용하였으며, 퓌레를 제조한 후 품질특성과 저장성을 조사하였다. 돼지 뒷다리육과 소, 돼지 혀로 제조된 퓌레는 단백질 함량이 10%이상으로 높고, 철 함량 등 무기물 함량과 필수아미노산 조성이 좋아 어린이들에게 유용한 영양소를 공급할 수 있을 것으로 판단되며, 또한 10℃에서 15일 이상 저장이 가능하기 때문에 최소 유통기간은 확보될 것으로 생각된다.
< 천연소재 조사, 탐색 및 이용효과 구명>
○ 천연첨가소재선발을 위한 소재로써 천연보존료 및 항산화제로는 mustard, marjoram, chrysanthemum, oregano, sage, thyme, rosemary, green tea 등의 추출물 또는 분말을 사용하였음. 천연색소 첨가 소재로는 안토시아닌(anthocyanin)이 주성분인 자색고구마(purple sweet potato), 적양배추(rad cabbage), 베리류(berries) 추출색소 등이며, capsanthin이 주성분인 파프리카(oleoresin paprika), 캠시컴(Capsicum) 추출색소 등을 사용하였음. 또한 조직감 향상 천연소 재로는 쌀겨(rice hull), 콩 껍질(soy hull), 삶은 우거지(boiled radish leaves), 삶은 배추(boiled Korean cabbage), 귤껍질(orange peel), 사과찌꺼기(apple pomace) 등에서 추출한 식이섬유를 활용하였음. 패티 에 천연첨가물을 다양한 농도로 첨가한 후 제조한 다음 가공저장과정에서의 이화학검사, 미생물검사 및 관능검사를 통하여 천연발색, 항산화, 항균, 조직감 개선 등의 기능성을 분석하여 최종적으로 oleoresin paprika추출물, green tea추출물, sage추출물을 선발함.
○ 천연 첨가제의 첨가수준이 높아질수록 패티 저장중의 항산화, 항균 및 색상에 대한 기능성이 증가하였으나 sage나 green tea 추출물의 경우 첨가수준이 높아질수록 관능적인 측면에서는 낮게 나타났음. 반면 oleoresin paprika추출물은 첨가수준이 높아질수록 관능적인 측면에서는 높게 나타났음. 따라서 선발된 천연첨가소재의 육제품 내 배합비의 최적화를 위한 첨가수준설정에서 흥미구역으로서 sage추출물, green tea추출물 및 oleoresin paprika추출물의 첨가수준을 총 원료대비 최대 0.5%(w/w)로 조절하였음.
○ 선발된 천연소재에 대한 첨가수준의 최적화를 위하여 심플렉스 격자설계를 이용한 혼합물설계 법을 이용하여 최적화를 이루었으며 프레스햄의 경우 최적점(최적첨가농도)을 분석한 결과 총 원료대비 oleoresin paprika추출물 0.28%(w/w), sage추출물 0%(w/w), green tea추출물 0.22%(w/w)로 계산되었음. 본 최적점에서의 종합만족도(overall desirability)는 0.952로 높게 나타났음. 소시지의 경우 총 원료대비 oleoresin paprika추출물 0.25%(w/w), sage추출물 0%(w/w), green tea추출물 0.25%(w/w)로 계산되었으며 종합만족도(overall desirability)는 0.884로 나타났음. 패티의 경우 총 원료대비 oleoresin paprika추출물 0.323%(w/w), sage추출물 0.077%(w/w), green tea추출물 0.10%(w/w)로 계산되었으며 종합만족도(overall desirability)는 0.734로 나타났음.
○ 저장성실험에서 대조구와 천연소재 첨가 프레스햄과 소시지 모두 4℃ 저장 중 28일 동안에 pH와 TBARS값의 큰 변화가 감지되지 않았음. 아질산염을 첨가한 대조구가 천연소재 첨가 군에 비하여 저장 중 총균수의 증식속도에서 상대적으로 크다는 것을 알 수 있었으며 천연소재의 항균작용을 감지할 수 있었음. 패티의 경우 TBARS 변화로 28일 동안에 대조구는 318% 증가하였으나 green tea추출물의 배합률이 상대적으로 높게 제조된 패티의 경우에는 거의 증가되지 않았음.
○ 천연소재로서 항산화, 항염 및 항혈전 작용의 기능성을 지닌 도인, 둥근마, 모과, 목단피, 물오리나무, 석창포, 오미자, 자완, 조팝나무, 참가시나무, 토복령 등의 에탄올추출물에 대한 대식세포를 이용한 독성여부, 항산화, 항염 등을 비교분석하여 심혈관계 질환예방 육가공제품소재로 모과를 선발하였음.
○ 모과추출물의 농도(w/w)가 증가할수록 관능적기호도는 떨어졌으며 0.5%농도의 수준에서는 전반적인 평가에서 대조구와 유의성차이가 나타나지 않았으므로 천연육제품(패티, 소시지, 햄)에 0.5%를 모과첨가수준으로 설정함.
○ 모과추출물을 첨가한 개발 제품(패티, 소시지, 햄)의 저장과정에서 모과를 미세캡슐화한후 첨가한 경우 이화학검사, 관능검사 및 미생물검사를 통하여 품질특성을 분석한 결과 비캡슐화 제품보다 4℃ 저장 중 7-35일 동안 저장과정에서 기능성이 상대적으로 높게 나타났음.
< 천연소재의 캡슐화공정개발 및 천연소재와 개발된 육가공 제품의 기능성 연구>
○ 미세캡슐공정의 최적공정조건으로 green tea와 sage 추출물을 위한 미세캡슐화의 피복물질 로는 cyclodextrin : maltodextrin 비율을 1 : 3으로 혼합 배합하는 것이었으며 oleoresin paprika 추출물을 위한 피복물질로는 maltodextrin을 단독 사용하는 것이었음.
○ 가공적성(열처리공정조건)에 따른 캡슐하지 않은 것과 미세캡슐화한 green tea와 sage추출물의 총 폴리페놀 함량, 총 플라보노이드 함량, hydroxyl radical 소거능 등을 비교분석한 결과 가열온도가 높아지고 가열시간이 길어짐에 따라서 함량 및 기능성이 줄어들었음. 열처리과정에서 미세캡슐화한 추출물의 경우 캡슐화하지 않은 것에 비하여 감소속도가 월등히 작았음. 예로서 고압살균공정인 120℃에서 60분간 열처리 시 캡슐화하지 않은 green tea추출물과 미세캡슐화한 green tea추출물의 총 플라보노이드 함량은 각각 23.6%와 4.8% 수준의 감소가 나타났음. 열처리공정조건에 관계없이 색도변화에서도 미세캡슐화한 추출물의 경우 캡슐화하지 않은 것에 비하여 색도에 대한 변화가 적었음.
○ 저장안정성비교에서 미세캡슐화한 green tea와 sage 추출물이 저장조건에 관계없이 저장과정에서 캡슐화 하지 않은 시료에 비하여 총 폴리페놀과 총 플라보노이드 보존효과가 컸음. 예로서 4℃의 50% RH의 저장조건에서 24일 저장 후 캡슐화 하지 않은 green tea추출물은 총 플라보노이드 함량이 19.7% 감소가 나타났으나 미세캡슐화한 경우는 거의 감소하지 않았음. 미세캡슐화한 oleoresin paprika추출물의 경우는 저장조건에 관계없이 저장 24일 기간 내내 캡슐화 하지 않은 것에 비하여 높은 적색도(a값)를 보였음.
○ 미세캡슐화한 green tea, sage, oleoresin paprika 추출물(처리구 2), 비 캡슐의 green tea, sage, oleoresin paprika 추출물(처리구 1) 및 아질산염이 첨가된 대조구를 패티, 소시지 및 프레스 햄 등의 육가공제품에 적용하여 가공적성(열처리)을 비교분석한 결과 소시지와 프레스 햄의 경우 가열전과 후 모두 DPPH radical 소거활성은 처리구 2가 가장 높게 나타났으며 그다음 처리구 1, 대조구 순이었음. 소시지의 경우 가열 후 대조구의 DPPH radical 소거활성은 처리구 2의 30.7%수준이었음.
○ 저장안정성분석에서 패티의 경우 4℃저장 7일후에 처리구 1과 처리구 2는 DPPH radical 소거활성이 각각 12.74와 18.45%이었으나 대조구의 경우에는 6.63%수준에 불과하였음. 처리군간에 비교로써 미세캡슐화(처리구 2)가 비 캡슐화(처리구 1)에 비하여 DPPH radical 소거활성에 도움이 크게 되는 것으로 나타났음. 소시지와 프레스 햄의 경우서도 4℃저장 35일 후에 DPPH radical 소거활성이 처리구 2가 가장 높았으며 처리구 1, 대조구 순 으로 나타났음.
< 천연소재의 캡슐화공정개발 및 천연소재와 개발된 육가공 제품의 기능성 연구>
○ 녹차, 자몽종자추출물, 코치닐 첨가시 농도에 의존적으로 혈전용해능이 증가하였으며, 자몽종자추출물의 혈전용해능이 유의하게 가장 우수하였다.
○ 녹차추출물 및 코치닐을 캡슐화한 시료는 일반추출물에 비해 혈전용해능이 증가하는 경향이었고, 캡슐화 자몽종자추출물은 일반자몽종자추출물에 비해 혈전용해능이 유의하게 증가하였다.
○ 정상쥐 혈액에서 혈액응고시간은 녹차>코치닐>자몽종자추출물순으로 증가하였으며, 자몽 종자추출물과 코치닐의 캡슐화 시료의 혈액응고지연효과가 우수하였음. 고혈압쥐에서는 캡슐화 녹차추출물의 혈액응고시간이 녹차추출물에 비해 증가하였다.
○ 항염증실험에서 자몽종자추출물은 100ug/mL 농도에서 세포독성을 나타냈으나, 녹차 및 코치닐은 동일한 농도에서 세포독성을 나타내지 않았음. 녹차, 자몽종자추출물은 NO, 지질과산화물 생성을 억제, iNOS 단백질 발현을 억제하였고 자몽종자추출물의 억제효과가 가장 컸다.
○ 세이지추출물의 미세캡슐화추출물은 혈전용해, 혈액응고지연을 시키는 효과가 우수하였으며, murine RAW 264.7 대식세포주를 이용하여 항염증반응 효과를 평가하였을 때 미세캡슐화 세이지 추출물이 NO 생성을 억제하고 지질과산화물 생성을 억제하였다.
○ 고과당 식이는 체중증가를 유발하였으나 녹차추출물과 미세캡슐화 녹차추출물 섭취시 체중증가를 억제하였으며 미세캡슐화 녹차추출물 섭취군에서 가장 체중증가가 유의하게 낮았다.
○ 고과당 식이를 섭취시 혈중 중성지질농도 및 유리지방산 농도가 유의하게 증가하였는 데 녹차추출물 섭취시 혈중 중성지질농도가 감소하였으며, 미세캡슐화 녹차추출물섭취시 가장 낮았다.
○ 고과당 식이로 인해 혈중 콜레스테롤이 증가하였으나 녹차추출물섭취시 혈중 콜레스테롤이 유의하게 감소하였다.
○ 간, 내장지방, 부고환지방의 지방합성효소인 malic enzyme 활성은 고과당식이로 인해 증가하였는 데 녹차추출물과 미세캡슐화녹차추출물 섭취시 유의하게 감소하였으며, 미세캡슐화의 상승효과는 없었다.
○ 고과당식이로 인해 증가된 심장조직내 섬유화를 녹차추출물이 억제하였으며, 미세캡슐화 시 그 효과가 상승하였다.
○ 따라서 in vivo 실험동물모델에서 녹차추출물은 혈중 중성지방, 콜레스테롤을 감소시키고 내장지방 축적을 억제하였다. 미세캡슐화 추출물의 섭취는 일반추출물에 비해 혈중 중성지방 감소, 혈압감소, 혈중 유리지방산농도 감소, 내장지방축적억제에 효과적이어서 지질대사를 개선하고 혈행기능개선효과를 증진시킬 수 있었으며 심장조직내 섬유화를 억제하였음을 알 수 있었다.
○ 모과추출물 또는 미세캡슐화 모과추출물을 첨가한 육류패티 섭취시 체중증가, 혈중 중성 지방, 혈중 콜레스테롤, HDL 콜레스테롤은 각 실험군에서 유의적인 차이가 없었다.
○ 육류패티섭취군에 비해 모과추출물 첨가 육류패티 섭취군의 내장지방 무게가 가장 적었다.
○ 간에서 고과당 육류패티 섭취군의 catalse 활성이 감소하였으나 미세캡슐화 모과추출물 섭취군에서 catalase 활성이 가장 높았다.
○ 고과당 육류패티 섭취시 부고환지방, 내장지방에서 지방합성 효소 활성이 증가하였는 데 모과추출물 첨가 패티 섭취군에서 부고환지방의 malic enzyme, 내장지방의 G6PD 활성이 유의하게 낮았으며, 미세캡슐화 첨가 육류패티 섭취군의 활성이 가장 낮았다.

Innovation and development of manufacture technologies for the meat processed products specially for children to enhance digestibility and absorbability as well as convenience to consume, are the important issues of food processors. In the present study we have performed several trial manufactures of meat food products and their chemical composition, technological quality and sensory characteristics are summarized below.
1. Development of absorption/digestibility enhancement meat products using coating technology with natural substance The objective of this study was to develop a manufacture technology of high proteolytic enzymes fruit peels - containing meat products to improve protein absorption when being consumed by children. For the setting of manufacture technology, 4 kinds of fruit peels (kiwi, pineapple, pear and fig) coated with sodium chloride and sodium aliginate were incorporated into meat products. The outcome of our analysis shows that the additions of coated kiwi and pineapple feels resulted in improvements of sensory properties and texture traits (e.g., hardness and springiness). In addition, there were no significant differences in physicochemical, texture and sensory properties among the treatment and control groups during 30 days of refrigerated storage. The effects of fruit peel coatings on protein digestibility were studied in rat. Our results revealed that no protein digestibility improvement effects were observed for the coated fruit peel hams – fed rats. However, the rates of body weight gains of rats in the treatment groups were significantly higher than that in the control group under the same feeding period. These suggest that ingestion of meat product containing coated fruit peels may help to supplement the protein source needed in children who have protein digestion disorder.
2. Development of meat products for child nutrient deficiency using natural substance
Mineral and vitamin contents are very important for human health maintenance, especially for children. It has long been known that liver of animal is the rich sources of essential nutrients. In this study, we have designed several manufacture methods of meat products with incorporations of cattle and pig livers, and the physicochemical composition, technological quality and sensory traits of the products were also evaluated during storage. Our results show that sausage made with heated liver possessed higher sensory scores than the sausage made with raw liver. Furthermore, 50% of the amount of fat in the meat products were replaced by heated liver and the quality and self-life stability of the liver sausages were investigated. The results show that there were no significant differences in physicochemical composition, texture attributes, sensory properties and microbial change among treatments. The preference score was decreased by the additions of cattle and pig livers. TBARS and VBN values in liver sausage groups showed higher as compared with the control group.
3. Development of meat products with high convenience for children consumption
This study was conducted to manufacture meat products which children can easily eat.
Puree was prepared with pork hind parts and cattle tongue and pig tongue respectively.
The outcome of our analysis shows that the purees prepared with pork hind parts, cattle tongue and tongue pig had high protein content (more than 10%). The mineral content and essential amino acids composition were superior in the purees made with pork hind parts and cattle tongue and pork tongue respectively. Therefore, we assume that these products could be used to supply the useful nutrients for children. In addition, these products were found had a minimum shelf-life stability of more than 15 days at 10℃.
< Investigation of survey, search and utilization effects for natural materials>
Meat industry uses different kinds of preservatives, yet those most frequently applied are nitrites and nitrates, which serve as curing agents in meat, not only as preservatives, but also for colour and flavor formation. However, sodium nitrite plays also a considerable role in the formation of carcinogenic N-nitrosamines. Therefore, meat industry is responsible for the delivery of safe products to the market, and recently, it tends to limit the amounts of nitrites and nitrates added to processed meat by replacement of these compounds with other substances such as natural pigments, preservatives and antioxidant.
This research was carried out to investigate and select natural pigments, antioxidant, antibiotics and texture improvements additives, and to determine maximum concentration and optimize mix proportioning of selected natural additives for substitutes of nitrite or nitrate, and to develop new meat products such as patties, sausages and press hams for prevention of cardiovascular disorders.
Extracts or powders of mustard, marjoram, chrysanthemum, oregano, sage, thyme, rosemary and green tea were used for natural preservatives and/or antioxidant of meat products. Purple sweet potato, rad cabbage, berries oleoresin paprika and capsicum extracts were used for natural pigments. Extracted dietary fiber from rice hull, soy hull, boiled radish leaves, boiled Korean cabbage, orange peel and apple pomace were used for texture improvements.
Functional properties of coloring agent, antioxidant and antibiotics for meat products such as patties, sausages and press hams were increased during storage as increasing of selected additives, oleoresin paprika, green tea and sage extracts concentrations. While, limited concentration of natural additives was 0.5%(w/w) based on sensory hedonic test results.
Oleoresin paprika, green tea and sage extracts additions as natural additives based on simplex lattice composite design were applied to analyze the quality of patties, sausages and press hams using response surface methodology. The optimal mix proportioning of oleoresin paprika, sage, green tea and overall desirability by analysis of multiple response surface methodology with the same weighed value of each dependent variable were 0.28%, 0%, 0.22% and 0.952 for press ham, respectively. The optimal mix proportioning of those were 0.25%, 0%, 0.25% and 0.884 for sausage, and 0.32%, 0.08%, 0.10% and 0.724 for patty, respectively.
The values of pH and TBARS of control(added 0.01% NaNO2) and treatment(added 0.5% natural additives) were almost constant during 28 days of storage for press ham and sausage at 4℃. Total plate count of control was higher than those of treatment, which meant that natural additives had more antibiotic property than control.
Ethanol extracts of P runes persica S., D ioscortea opposita T., Chaenomeles sinensis K., M outan cortex R., Alnus hirsuta, Acorus gramineus S., Schizandra chinensis, Aster tataricus, Spiraea prunifolia S., Quercus salicina and Smilax china L. were used to investigate and select natural additive for prevention of cardiovascular disorders, and to develop new meat products such as patties, sausages and press hams. Quince(Chaenomeles sinensis K.) was selected based on toxicity using macrophage test, antioxidant and anti-inflammatory results.
Limited concentration of quince for patties, sausages and press hams was 0.5%(w/w) based on sensory hedonic test results, which had no significant difference in comparison with control at 5% level.
The micro-encapsulation of quince had higher functional properties such as polyphenol and total flavonoid retentions, coloring agent, antioxidant and antibiotic for patties, sausages and press hams than non-capsulated quince during 7-35 days of storage at 4℃.
< Study of encapsulation process development and functional property for developed meat products with natural material>
Micro-encapsulation defined as a process to entrap bioactive agent(core material) within another substance(wall material). The micro-encapsulation aims to preserve stability of the bioactive compounds during processing and storage and to prevent undesirable interactions with food matrix. Mainly, bioactive compounds are characterized by rapid inactivation. These compounds would profit from an micro-encapsulation procedure, since it slows down the degradation processes(e.g., oxidation or hydrolysis) or prevents degradation until the product is delivered at the desired sites. Thus, the bioactive component would be kept as fully functional.
Also, this technology may provide barriers between sensitive bioactive materials and the environment.
Optimum processing conditions of micro-encapsulation were analyzed by yield, total polyphenol contents, SOD-like activity, total antioxidant activity and DPPH radical scavenging activity. The proportion of cyclodextrin to maltodextrin as wall materials was 1 : 3 for green tea and sage extracts as core materials. Maltodextrin was wall material for oleoresin paprika extract as single wall material.
For the processing aptitude(heat treatment processing) comparison of micro-encapsulated bioactive compounds such as green tea, sage and oleoresin paprika extracts(treatment) with non-capsulated bioactive compounds(control), Total polyphenol contents, total flavonoid contents and hydroxyl radical scavenging activities of treatment and control were decreased as the increasing of heating temperature and time. While, those of treatment had higher retention rate than those of control by comparison before with after heat processing. For example, total flavonoid contents loss rate of treatment and control comparison of after 60 min of heat processing at 120℃ were 4.8 and 23.6%, respectively. Also, color retention rate of treatment had higher value than that of control after heat processing.
For storage stability of total polyphenol contents and total flavonoid contents of green tea and sage extracts, those of treatment had higher retention rate than those of control without storage conditions. For example, total flavonoid content of green tea extract were not changed after 24 days with 50% RH, 4℃ storage conditions. While, that of control decreased 19.7%. Color redness(a) of treatment for oleoresin paprika extract had higher value than that of control throughout 24 days with 50% RH, 4℃ storage conditions. Bioactive compounds(green tea, sage and oleoresin paprika extracts) treated with micro-encapsulation as treatment 2, non-encapsulation as treatment 1 and added NaNO2 as control were applied to meat products such as patty, sausage and press ham for the analyzing of processing aptitude. DPPH radical scavenging activity had the highest value in treatment 2, and then treatment 1 and control after heat treatment for sausage and press ham. The DPPH radical scavenging activity of control was only 30.7% compared to that of treatment 2 after heat processing in the case of sausage and press ham.
DPPH radical scavenging activities of treatment 1, treatment 2 and control for patties were 12.74, 18.45 and 6.63% after 7 days storage at 4℃, respectively, which meant that micro-encapsulation techniques provided barriers between sensitive bioactive materials and the environment. DPPH radical scavenging activity had the highest value in treatment 2, and then treatment 1 and control after 35 days of storage 4℃ for sausage and press ham.
In vitro, in vivo bioassay of bioactive materials for improving functionality of meat products The aim of this study was to compare the effects of extracts from natural food materials and their encapsulated extracts on cardiovascular diseases using in vitro and in vivo model.
In 1st year, we compared the effects of grapefruit seed extract(GFSE), green tea extract(GTE), cochineal and their microencapsulated extract on anti coagulaion, fibrinolysis, cell cytotoxicity and anti-inflammatory activites in murine RAW264.7 macrophages cell line. Nitric oxide(NO), prostaglandin E2(PGE2), tumor necrosis factor-α(TNF-α), inducible nitric oxide synthase(iNOS) protein expression and thiobarbiturate reactive substances (TBARS ) were analyzed. In 2nd year, the effect of green tea extract (GTE) and its microencapsulated green tea extracts on blood lipid profiles and cardiovascular system was evaluated using hypertriglyceridemic animal model. the bioactive component from green tea extract (GTE) rats were administrated orally (50 mg/kg body weight/day) as free or in a microencapsulated form with maltodextrin in rats fed a high fructose diet. In 3rd year, we compare the effect of quince extract and its microencapsulated extract containing meat patty on blood lipid profile and cardiovascular system in hypertriglyceridemic rat model.
1. The addition of GTE, GFSE ,or cochineal increased fibrinolysis in dose dependent manner and the grapefruit seed extract had the significant effect on fibrinolysis compared to green tea extract or cochineal.
2. The microencapsulation of GFSE increased fibrinolysis further than the non encapsulated GFSE significantly.
3. The blood coagulation time was extended by the addition of microencapsulated GFSE or microencapsulated cochineal in normotensive rats. In spontaneously hypertensive rats, the blood coagulation time was extended by the addition of microencapsulated GTE.
4. We observed cell cytotoxicity after adding GFSE with dose of 100ug/mL in murine RAW264.7 macrophages cell. The green tea extract at the range of 100-600㎍/mL that inhibited NO and PGE2 production and iNOS protein expression without cytotoxicity in dose dependent manner. Grapefruit seed extract had strong inhibitory effects on NO and PGE production and iNOS protein expression at the range of 5-20㎍/m. Microencapsulation of green tea extract had futher inhibitory effects on NO and PGE2 production and iNOS protein expression while microencapsulated GFSE did not show further inhibitory effect on these parameters.
5. High fructose diet induced features of metabolic syndrome including hypertriglyceridemia, hyperuricemia, increased serum total cholesterol, and retroperitoneal obesity. In addition, myocardial fibrosis was increased.
6. In rats receiving high fructose diet, the lowering of blood triglycerides, total cholesterol, non esterified fatty acid and uric acid, as well as the reduction in final body weight and visceral fat weight associated with the administration of GTE, led to a reversal of the features of metabolic syndrome (p < 0.05).
7. In particular, the administration of microencapsulated GTE decreased myocardial fibrosis and increased liver catalase activity with a further alleviation of serum NEFA, and hyperuricemia compared to administration of GTE.
8. The intake of meat patty diet containing quince extract or microencapsulated quince extract did not affect the final body weight, triglyceride, total cholesterol, HDL cholesterol in hypertriglyceridemic rats compared to meat patty diet only.
9. Feeding the meat patty diet with quince extract decreased visceral fat pad mass compared to feeding the meat patty.
10. The activity of catalase was lower in liver of meat patty and high fructose fed rats and the supplementation of microencapsulated quince extract increased catalase activity significantly.
11. The activities of lipogenic enzyme in epididymal fat and visceral fat tissue of high fructose and meat patty fed rats were higher compared to rats fed high fructose and meat patty containing quince extract diet. The lipogenic enzyme activity was decreased further by microencapsulation of quince extract.
Taken together, our results suggest that GSFE might be a promising candidate for preventing inflammation related diseases such as cardiovascular disease, cancer or diabetes and microencapsulation of green tea extract and quince extract could improve their bioactivities. In addition, the microencapsulation of the bioactive components of food materials might have protective effect on cardiovasucular system by attenuating the adverse features of myocardial fibrosis, and decreasing uric acid level and increasing hepatic catalase activity more effectively by protecting their bioactivities.
