평균수명의 연장은 노화와 관련된 기초연구와 이를 이용한 항노화 및 뇌질환 개선제 개발을 촉진하고 있다. 연구자체의 중요성과 개발결과가 국민보건에 미치는 영향을 고려해 볼 때 매우 중요한 과제이다. 녹차에 대한 최근의 연구에 의하면 in vitro, in vivo에서 항암, 항산화 등의 효능이 있는 것으로 밝혀졌으며, 임상적인 변에서 녹차의 가장 중요한 성분은 카페인과 플라보노이드이며, 이중 플라보노이드는 catecholamines 의 수준 증가작용, 말초혈관 강화작용, 항염증 작용, 갑상선 기능조절, 방사선에 대한 보호작용, 항균작용 등이 보고되어 있다. 그 외에도 GABA성분, 아미노산 중의 하나인 theanine 등도 녹차의 유효한 성 분 중의 하나로 주목받고 있다. 노화의 기전 및 노인성 질환의 치료제개발에 있어서 가장 큰 장애가 되었던 동물모델 (노화 촉진모댈 마우스, SAM) 이 개발됨에 따라 노화의 기전 및 노인성질환 연구에 박차를 가하게 되었다. 본 연구는 항노화 및 뇌질환 개선물질 개발을 위하여 SAMP8 (학습과 기억 장애)의 모델 동물로서의 정착과 녹차에 대한 약효검색(한국화학연구소), 녹차로부터의 약리 활성물질 탐색(태평양기술연구원), 녹차에 대한 독성병리시험(전북대학교)을 수행하였다. 녹차의 장기 투여가 뇌와 면역기능에 미치는 영향을 알아보고자, 노화 모델동물인 SAMP8과 SAMR1(SAMP8의 정상적인 대조군)의 암수 마우스를 녹차 추출물 0.2%와 1.0% 투여군 (T1 and T2) 및 투여하지 않은 대조군으로 나누어 생후 3개월부터 음수시켜 6개월령과 12 개월령 되었을 때 다음과 같은 결과를 얻었다. SMAR1 과 SMAP8 마우스의 각 실험군간에 뇌의 육안적 관찰에서 차이점이나 병변이 발견되지 않았다. 그러나 뇌의 비교중량치는 녹차투여군이 대조군보다 현저한 증가치를 나타내었다. 또한, 대조군에서 볼 수 있었던 대뇌의 neuron과 axon의 퇴행성 변화와 수종성 변화가 양 strain의 녹차 투여군에서는 거의 발견되지 않은 것으로 보아 녹차는 대뇌의 노화성 퇴행성 변화를 억제하는 효과가 있는 것으로 사료된다. 뇌손상에 대한 biomaker라 할 수 있는 GFAP를 분석한 결과, SAMR1 과 SAMP8 사이에 유의한 차이가 있었으며, 녹차 투여농도와 상관성 있게 GFAP의 함량이 감소하여 중추신경계 개선의 효과가 입증되었다. 한편, 녹차의 주요성분인 카테킨과 GABA 성분을 증강시킨 GABA차를 28 일간 경구투여한 후 행동실험을 수행한 결과 단기기억 능력이 향상되었다. 면역계의 대표적 장기인 비장에 대한 실험에서도 비장 면역계조직세포의 퇴행성 변화가 양 strain의 녹차 투여군에서는 거의 발견되지 않은 것으로 보아 녹차는 비장 변역계세포의 퇴행성 변화를 억제하는 효과가 있는 것으로 사료된다. 한편, 녹차의 유효성분에 대하여 랫드를 이용한 약물동태실험 결과 카페인과 데아닌을 동시에 투여할 때 카페인의 흡수속도는 빨라졌으나 유의성은 없으며, 분포, 대사 및 배설에는 영향을 미치지 않았다. 카테킨과 카페인을 동시 투여하면 카페인의 흡수속도는 약간 빨라졌으나 카페인과 theobromine, theophylline, paraxanthine의 뇨중 배설량 (카페인 투여량의 6.6% vs 9.3%) 과 혈중 농도는 카페인 단독 투여시보다 감소하였으므로 카테킨은 동시 투여시 위내에서 카페인의 흡수량을 감소시키지만 흡수 속도에는 영향을 주지 않는다고 보여진다. 녹차 플라보노이드 성분을 분석한 결과 4종류의 주성분 플라보노이드는 (- )-epicatechin, (-)-epigallocatechin gallate, (-)-epigallocatechin, (-)-epicatechin gallate 등으로 동정 되었다. 이를 바탕으로 그동안 단편적으로 항산화 효과 등이 연구되어 온 녹차의 플라보노아드 성분의 의약품화 가능성을 타진하고자 각각의 플라보노이드 성분을 분리 추출한 결과 (-)-epigallocatechin gallate(61%), (- )-epicatechin gallate(12%) , (-)-epigallo catechin(15%), (-)-epicatechin(6%) 으로 그 조성이 밝혀졌다. GABA 성분은 녹차를 혐기적 조건으로 처리함에 따라 혐기 처리하지 않은 녹차엽에 비하여 GABA와 alanine 의 함량은 4-10배까지 현저하게 증가한 반면 glutamic acid는 현저하게 감소하였다. 이를 이용하여 GABA 함량을 증가시킨 GABA 차를 제조하였다. 또한 녹차의 아미노산 성분을 분석하고 L-theanine을 분리하였으며, 특히 폴리페놀 함량이 높은 녹차 추출물의 제조 방법(특허출원 제 62755호)을 확립하였다. 이상과 같이 본 연구에서는 노화촉진 마우스를 이용하여 녹차의 추출물과 주요성분에 대한 항노화 효과를 입증하였으며, 농축카테킨 함유 [카테키나]를 개발하여 시판하고있으며, 향후 녹차추출물을 이용한 약국경로 건강식품 등 항노화 개선제의 개발에 기여할 것으로 예상된다.

The increase of life expectancy prompted the basic research of aging mechanism and the development of anti-aging agents. Recent research on green tea reveals that its abstract suppresses the development of cancer and according to clinical reports the most important elements of green tea are caffeine and flavonoids and these materials, especially flavonoids, have many beneficial functions such as increase of catecholamines in brain, strengthening capillary blood vessels, anti -inflammatory effects, protection from radioradiation, and suppression of pathogenic activity. Moreover, the increasing importance of minor elements of green tea, such as GABA and theanine, is evidently proved. The availability of animal models for aging research has made it possible to study the basic mechanisms of aging and develop anti-aging reagents. This research is made up of three areas; the establishment of SAMPS as animal model for aging research and basic research on aging parameters for this type of research (Korea Research Institute of Chemical Technology), the search for "drugable substances" from green tea abstracts (Pacific Corporation!R&D Center) and neuropathogenic study of green tea abstracts (Chonbuk National University). In order find out the long-term effects of green tea administration, we orally dosed green tea abstract (0.2%, T1 and 1.0%, T2) into SAMPS and SAMR1 (the control strain of SAMPS) for 3 and 6 months starting at theage of 3 months. No obvious neuropathological effects were not detected in both strains of treated and untreated groups. However, green tea-treated group showed heavier brain weights than untreated control. In addition, apoptotic changes in neuron and netronal axon in SAMPS was not detected in green tea-treated group, implying that green tea is effective in general suppression of chronic symptoms of brain aging. The contents of GF AP, the biomarker of brain demage, were lower in SAMR1 than SAMPS, and green tea-treated group than untreated control groups, suggesting that green tea has beneficial effects in brain damage resulted from agmg. Green tea constituents, catechin and GAB A, also showed increased short-term memory when animals were orally administered with "GABA tea" for 2S days. In immune system, green tea treatment suppressed the splenic regression in both strains. The parmacokinetics of theanine and caffeine in SD rats were not obviously different in treated and vehicle-treated groups. We found that the flavonoids of green tea are composed of (-)-epigallocatechin gallate(61%), (-)-epicatechin gallate(12%), (-)-epigallo catechin05%), and (-)-epicatechin(6%). We also found that green tea treated in anaerobic condition contained 4-10 times more GABA and alanine; the contents of glutamic acids were significantly decreased. This information was used to produce "GABA tea" which contain more GABA in the tea mixture. We also established method for preparation of green tea abstract containing high polyphenol and this method was patented (Patent number, 62755). In summary, we established the effects of green tea and its constituents in aging using animal model, and this information was applied to produce anti-aging reagents.
