본 연구는 식품 중 프로필렌글리콜 분석법 개선 및 검증, 국내유통 식품 중 프로필렌글리콜 함량 분석 및 식품섭취량 자료를 통해 우리나라 프로필렌글리콜 섭취량을 평가하였다.

본 연구는 식품 중 프로필렌글리콜 분석법 개선 및 검증, 국내유통 식품 중 프로필렌글리콜 함량 분석 및 식품섭취량 자료를 통해 우리나라 프로필렌글리콜 섭취량을 평가하였다.
식품 중 프로필렌글리콜 함량 분석을 위해 「식품 중 식품첨가물 분석법」수록 분석법을 검토한 결과, 시료 전처리 조건에서 다양한 식품에 적용이 가능하도록 추출용매를 물:아세톤(30:70)으로 사용하고 지방 및 단백질 침전을 위해 카레즈 시액 I 및 II를 사용한 후 원심 분리하는 과정으로 개선하였다. 또한, 기기분석 조건에서 컬럼을 피크 비대칭이 심한 Agilent HP-5 column(0.25 μm, 30×0.25 mm)에서 Agilent INNOWAX column(0.25 μm, 30×0.25 mm)으로 변경하고 승온 조건 등을 변경하여 분석법을 확립하였다. 분석법 검증결과, 회수율은 약 93.0~97.6%, 정밀성(%RSD)은 0.9~6.5% 이하, 검출 및 정량한계는 각각 0.39 mg/kg, 1.32 mg/kg, 직선성(R2)은 1~100 mg/L에서 0.999로 우수하였다. 또한, 만두 등 4개 식품에 대해 측정불확도를 산출하였다.
국내유통 식품 중 프로필렌글리콜 함량 조사를 위해 프로필렌글리콜이 함유될 가능성이 있는 식품(1,073건)을 구입하여 분석하였다. 분석한 시료는 모두 식품첨가물공전의 사용기준에 적합하였으며 검출범위는 불검출~1.3%(캔디류)이었다.
식품을 통한 프로필렌글리콜 섭취량 평가를 위해 식품 중 프로필렌글리콜 함량 결과와 최근 3년간 국민건강영양조사(2011~2013) 자료의 식품섭취량을 적용하여 프로필렌글리콜의 일일추정섭취량을 산출하였으며, 프로필렌글리콜 일일섭취허용량(ADI) 대비 섭취 수준을 평가하였다. 식품을 통한 프로필렌글리콜 섭취량 평가 결과, 전체인구집단의 평균 프로필렌글리콜 일일섭취량은 26.3 mg/day, 남자는 29.1 mg/day, 여자는 23.6 mg/day이었으며, 일일섭취허용량(25 mg/kgbw/day) 대비 각각 2.1%, 2.1%, 2.0%로 안전한 수준이었다. 섭취자 집단의 평균 프로필렌글리콜 일일섭취량은 34.3 mg/day, 남자는 37.1 mg/day, 여자는 31.4 mg/day로 일일섭취허용량(25 mg/kgbw/day) 대비 각각 2.7% 수준이었다.
본 연구결과에서 식품 중 프로필렌글리콜 분석법 및 섭취량 평가 결과는 국내외적으로 학술적 가치가 있을 뿐 아니라 정책 활용 및 안전성에 대한 기초자료로 활용될 것으로 기대된다.

This study aimed to assess the daily intake of propylene glycol (PG) by the Korean population through the improvement of the method and analysis of PG in foods retailed in Korea.
The PG method in 「Analytical Method of Food Additives in Foods」 was changed to be applicable to determine PG in various foods. In sample preparation, PG in foods was extracted with water:acetone(30:70), followed by the treatment of Carrez solution I, II and centrifugation. The improved method was established through the change of Agilent HP-5 column(0.25 μm, 30×0.25 mm) to Agilent INNOWAX column(0.25 μm, 30×0.25 mm) and conditions such as temperature program and split ratio. The method validation showed that recoveries of PG ranged from about 93.0% to 97.6%, with relative standard deviation (RSDs) between 0.9% and 6.5%. Limit of detection and limit of quantification were 0.39 mg/kg, 1.32 mg/kg, respectively. Good correlation coefficients(r2>0.999) was showed in the ranges of 1~100 ppm. In addition, measurement uncertainties were calculated based on four food matrixes (dumpling, coffee, candy and chocolate).
The PG content of 1,073 products retailed in Korea was analyzed. All levels of PG were below the standard of「Food Additives Code」, and ranged from N.D. ~ 1.3%(candies).
The daily intake of PG was estimated based on the monitoring data and Korean National Health and Nutrition Examination Survey(KNHNES)(2011-2013) to evaluate the intake of PG in the Korean population. Mean PG intake by general population, male and female were 26.3 mg/day, 29.1 mg/day and 23.6 mg/day, respectively, and 2.1%, 2.1% and 2.0%, respectively, compared with ADI (25 mg/kgbw/day). Mean PG intake by consumer group, male and female were 34.3 mg/day, 37.1 mg/day and 31.4 mg/day, respectively, and 2.7%, respectively, compared with ADI (25 mg/kgbw/day).
These results indicate that PG intake by the Korean population and the improved method have academic value and is useful for PG policy and safety.
