스테리그마토시스틴(Sterigmatocystin, STC)은 Aspergillus 속 곰팡이가 생성하는 2차대사산물로 폴리케타이드 곰팡이독소이다. 일반적으로 아플라톡신과 동일한 생합성 경로를 거쳐 생성되며, 구조적으로 매우 유사하다. 스테리그마토시스틴은 곡류 및 곡류가공품에서 검출이 보고되고 있다.

본 연구사업은 백미을 포함한 곡류 중 스테리그마토시스틴 및 총 아플라톡신 안전관리를 위해 LC-MS/MS를 이용한 동시시험법을 검토하였다. 13C 동위원소를 이용한 내부표준액 방법, QuECHeRs 방법, 고체상추출칼럼(Solide Phase Extraction, ISOLUTE)과 면역친화성칼럼(Immunoaffinity column, IAC)를 이용한 시험방법을 검토하여 곡류 중 스테리그마토시스틴 및 총아플라톡신을 분석하기 위한 시험법으로 고체상추출칼럼을 이용한 시험법을 확립하였다. 균질화된 시료를 50% 아세토니트릴로 추출 및 여과하고 SPE(ISOLUTE)를 이용해 정제한 후 액체크로마토그래피-질량분석기/질량분석기(LC-MS/MS)를 이용하여 분석하였다. 백미, 조, 수수, 옥수수, 밀가루 5품목에 대해서 특이성, 직선성, 검출한계, 정량한계, 매트릭스영향, 정밀성 및 정확성으로 시험법 유효성 검증을 수행하였다. 스테리그마토시스틴 및 아플라톡신 B1, B2, G1 및 G2의 검출한계 및 정량 한계는 각각 백미 0.03~0.99 ng/g, 수수 0.13~0.77 ng/g, 조 0.03~1.75 ng/g, 옥수수 0.03~0.48 ng/g, 밀가루 0.03~0.65 ng/g 수준이었다. 곡류 5품목의 평균 회수율은 68.66~119.84 % 이었으며, 상대표준편차는 15 % 이하로 양호한 결과를 얻었다. 각 곰팡이독소분석을 위한 matrix matched 검량선 곡선은 R2>0.999으로 직선성을 나타내었다. 백미 중 스테리그마토시스틴 및 총 아플라톡신류의 분석법에 대한 측정불확도는 GUM과 EURACHEM을 근거로 95% 신뢰수준에서 스테리그마토시스틴 및 AFB1, B2, G1, G2 각각 0.3 ± 0.03 μg/kg, 0.9 ± 0.09 μg/kg, 0.9 ± 0.26 μg/kg, 0.9 ± 0.12 μg/kg, 1.5 ± 0.33 μg/kg으로 산출되었다. 7개 실험실간 교차검증을 수행한 결과, 백미 및 수수 중 5종 곰팡이독소 회수율의 상태표준편차(RSDR)가 모두 12% 이하였다. 또한, 국내 유통 중인 곡류 3품목(백미, 조 및 수수) 총 60건을 구입하여 스테리그마토시스틴과 AFB1, B2, G1, G2를 분석한 결과, 조와 수수에서 스테리그마토시스틴과 AFB1, G2가 1 μg/kg이하의 미량으로 검출되었고, 수수 1건에서 스테리그마토시스틴과 AFB1가 동시에 검출되었다. 본 연구를 통하여 식품 중 곰팡이독소 안전 관리를 위해 유효성이 검증된 곡류 중 스테리그마토시스틴 및 총아플라톡신 동시시험법을 마련하였다.

The mycotoxin sterigmatocystin(STC) is one of polyketide mycotoxins as a secondary metabolite produced by Aspergillus fungi, a type of fillamentous fungi, and it has been reported as a contaminant in cereals and cereal products. Sterigmatocystin is generally produced by the same biosynthesis route with aflatoxins(AFs) production. Also, its structure is similar to that of aflatoxins. A test method is necessary to evaluate all stages of food production: from raw materials to the final products and even systematic monitoring of mycotoxin for food safety from the farm to the table.
This study examined and determined the simultaneous detection method using LC-MS/ MS for sterigmatocystin and total aflatoxins in cereals such as polished rice. Internal standard methods using 13C isotope, QuECHeRs, solid-phase extraction column (SPE, ISOLUTE) and Immune Affinity Column(IAC) method were reviewed and established the detection method using the solid phase extraction column for sterigmatocystin and total aflatoxins in cereals. The sample extracted and filtered by 50% acetonitrile in water and purified with SPE (ISOLUTE) was analyzed by liquid chromatography-mass/mass spectrometry(LC-MS/MS). The validation for method (specificity, linearity, detection limit, quantification limit, with precision and accuracy as recovery of the repeated five times for matrix effects and 3 concentrations) was carried out for 5 items: polished rice, millet, sorghum, maize and wheat flour. Limit of quantification for sterigmatoxystin and total aflatoxins B1, B2, G1 and G2 were 0.13~2.61 ng/g for polished rice, 0.13~0.77 ng/g for sorghum, 0.09~1.75 ng/g for millet, 0.09-0.42 ng/g for maize, 0.10-0.65 ng/g for wheat flour, respectively. The recovery test result was from 60.3 % to 119.8 %, and RSDR was under 15 % for mycotoxins in all 5 items. A matrix matched calibration curve for each of mycotoxin analysis was R2>0.9999 showing linearity. The measurement uncertainties in polished rice in terms of GUM and EURACHEM were 0.3 ± 0.03 ug/kg for sterigmatocystin 0.9 ± 0.09 ug/kg for aflatoxin B1, 0.9 ± 0.26 ug/kg for aflatoxin B2, and 0.9 ± 0.12 ug/kg for aflatoxin G1 and 1.5 ± 0.33 ug/kg for aflatoxin G2 at 95 % confidence level. According to the results of conducting cross validation among 7 inter-labs, RSDR for the recovery of 5 species of mycotoxins in polished rice and sorghum were all under 12 %. In addition, 60 cereal items (20 polished rices, 20 millets, and 20 sorghum) circulated in Korea were selected and monitored, and less than 1 ug/kg of sterigmatocystin, aflatoxin B1 and G2 were detected in millets and sorghums. 1 sorghum sample contained both sterigmatocystin and aflatoxin B1. This study established the simultaneous detection method of sterigmatocystin and total Aflatoxins in validated cereals for the safety management of aflatoxins in food.
