최근에는 각 규제기관의 약전 규격을 조화하여 규제지역마다 다른 시험법 및 기준을 적용해야 하는 제약업계의 부담을 덜어주고, 시험분석의 과학적 일관성 유지를 통한 시험결과 호환성 및 신뢰성 확보를 위하여 약전의 국제조화가 크게 증가하고 있다. 약전의 국제조화는 약전토론그룹(Pharmacopoeial Discussion Group, PDG)을 통해 지속적으로 확대되어 일반시험법 분야는 31 개 항목에 대해 이미 국제조화되었다.
대한민국약전 제11개정 일반시험법은 75 개로서 유럽약전(285 개), 미국약전(163 개), 일본약전(85개)에 비해 제공하고 있는 정보가 현저히 부족하다. 또한 지속적으로 확대되고 있는 국제조화 항목에 대한 검토 및 연구가 시급하다.
본 연구에서는 대한민국약전 일반시험법의 국제조화를 확대하고 제공하는 콘텐츠를 강화하기 위하여 대한민국약전 일반시험법 제·개정(안)을 마련하였다. 국내·외 약전 일반시험법을 비교 분석 및 현장 수요분석 등을 통해 선정 기준을 마련하였다. 국제조화 시험법 중 미반영 시험법, 약전 통합에 따른 舊)대한민국약전외의약품기준(KPC) 일반시험법, 글로벌 통용 시험법, 현장 수요 기반 시험법 등의 기준에 따라 겉보기밀도 및 탭밀도측정법 등 총 37 개 제·개정 대상 시험법에 대한 제·개정(안)을 마련하였다. 제·개정(안)의 완성도를 높이고 다양한 분야의 의견을 수렴하기 위하여 산·학·관 전문가협의체를 구성하여 운영하였고, 대한민국약전포럼을 2 회 발간하여 폭 넓은 의견수렴을 수행하였다. 아미노산시험법, 무기질시험법, 알코올수측정법 등은 실험을 통한 검증, 시험법 개선 및 밸리데이션을 수행하였다.
본 연구결과로 마련된 대한민국약전 일반시험법 제·개정(안)은 향후 대한민국약전에 반영되어 대한민국약전 국제조화와 제약업계 수출지원 및 품질관리 수준 향상에 크게 기여할 것으로 기대된다.

The 11th edition of the Korean Pharmacopoeia(KP 11) is the current official pharmacopoeia that has been designated by the Minister of Food and Drug Safety through an MFDS notice, following a review by the Central Pharmaceutical Affairs Council, as a means to ensure the appropriateness of the properties, conditions, quality, and storage methods of drugs. Countries around the world, including the U.S., EU member states and Japan, implement their national pharmacopoeias as a means to promote the quality of drugs.
Recently, many of the pharmacopoeias used by regulatory authorities have been harmonized for the purpose of easing the burden on pharmaceutical companies, which must implement various test methods and follow standards according to the region concerned. Also, the level of international harmonization of pharmacopoeias has increased considerably to maintain the scientific consistency of tests and analyses so as to ensure reliability and compatibility of the test results. The scope of international harmonization of pharmacopoeias has been expanded through the Pharmacopoeial Discussion Group (PDG), and of particular note, 31 items in the general test method category have become standardized to date.
KP 11, which specifies only 75 general test methods, provides substantially less information compared to the European Pharmacopoeia(285 general test methods), Unitesd States Pharmacopeia(163 general test methods), Japanese Pharmacopoeia (85 general test methods). Also, there is an urgent need to perform a review and analysis of the international harmonization items that are steadily growing in number.
In this study, a proposal for the establishment and revision of general test methods for KP was prepared for the purpose of enhancing the international harmonization of the general test methods set forth in KP and reinforce its contents. In addition, the selection criteria were determined based on the results of a comparative analysis of the general test methods prescribed in the domestic and foreign pharmacopoeias and a field demand analysis. The establishment and revision proposal includes a total of 37 test methods, including the apparent density and tapped density measurement methods that were chosen based on the criteria set forth: internationally harmonized test methods that have not yet been reflected in KP, general test methods prescribed in the former Korean Pharmacopoeia Codex (KPC), test methods that are globally applied, and test methods based on field demand. In order to enhance the integrity of the proposal and obtain input from a diversity of fields, an industrial-academic-governmental expert consultative group was formed and operated, and the KP Forum was issued twice to collect a wide range of opinions.
Then, empirical verification, test method improvement and validation were performed for the amino acid inorganic, and alcohol content determination test methods, among others.
The proposal for the establishment and revision of general test methods for KP (draft) formulated based on the results of this study is expected to be reflected in KP and significantly contribute to the international harmonization of KP, increase export for domestic pharmaceutical companies, and improve quality management.
