MIC test에 대한 결과 B. cereus, B. subtilis, E. coli, S. cholerae, S. flexneri, P. aeruginosa, S. typhimurium에서 MIC가 0.125 mg/㎖로 나타났고, L. monocytogenes, S. aureus, E. aerogenes 에서는 2배 높은 농도인 0.25 mg/㎖의 농도가 MIC로 나타났다. 효모에서는 Sacch. cerevisiae, C. utilis, D. hansenii 1 mg/㎖의 MIC가 나타났다. D. hansenii의 MIC와 MBC를 확인한 결과 MIC는 0.1 g/㎖, MBC는 0.15 g/㎖로 나타났고, 증식 및 생존억제 시험한 결과 0.15 mg/㎖로 항균물질을 첨가한 경우 D. hansenii는 60시간 동안 생존을 억제하였고, 0.1 mg/㎖의 경우는 D. hansenii의 증식을 12시간까지 억제를 보였다. 항균물질을 분리하기 위한 정제과정은 다음과 같이 수행하였다. In this study, lactic acid bacteria were isolated from kimchi for the study of non-protein antimicrobial compounds produced by lactic acid bacteria. Antibacterial activities were confirmed and selected. Selected lactic acid bacteria FC10 was identified as Lactobacillus plantarum as a result of 16S rRNA identification and was named Lb. plantarum FC10. In addition, the film yeast was directly isolated from the kimchi, and the 18S rRNA result showed 99% similarity with Debaryomyces hasenii. The antimicrobial activity of the antimicrobial compounds against the yeast was investigated. Separated lactobacillus Lb. plantarum FC10 did not show hemolysis, and antimicrobial compounds produced by Lb. plantarum FC10 was stable at pH and heat treatment, and was not decomposed by other enzymes such as Proteinase K, ALCALASE 0,6 L, Pancreatin. Therefore, it was judged that the antimicrobial substance was non-protein antimicrobial compounds. Also, It is smaller in size 100∼1,800 Da. UV scan result, the wavelength with the highest O.D. of the antimicrobial compounds was 245 nm. The antibacterial activity of Lb. plantarum FC10 was included in the top 10 of the 200 lactic acid bacteria in laboratory. As a result of the antimicrobial activity, B. subtilis, B. cereus, L. monocytogenes, S. aureus, E. aerogenes, E. coli, P. aeruginosa, S. typhimurium, Sacch. cerevisiae, C. utilis and D. hansenii showed inhibition spectrum. The results of MIC test showed that bacterial growth was inhibited at the concentration of 0.125 mg/㎕ in B. cereus, B. subtilis, E. coli, S. cholerae, S. flexneri, P. aeruginosa, S. typhimurium and inhibited at the concentration of 0.25 mg/㎕ in L. monocytogenes, S. aureus, E. aerogenes. In yeast, Sacch. cerevisiae, C. utilis, and D. hansenii was inhibited at the concentration of 1 mg/㎕. MIC and MBC about D. hansenii were found to be 0.1 g/㎖ and 0.15 g/㎖. When antimicrobial substances were added to 0.15 mg/ml, D. Hansenii caused no growth, and in 0.1 mg/ml, D. hansenii caused survival inhibition. Purification was performed to isolate the antimicrobial compounds. Lb. plantarum FC10 was cultured in CCM medium at 30℃ for 72 hours, and the supernatant was recovered and freeze-dried. After that, the supernatant which was freeze-dried was extracted from ethanol, heat-treated at 121℃ for 15 min with an autoclave and concentrated. After the heat treatment, gel filtration was performed to separate the antimicrobial compounds having different sizes, and purification was carried out by FPLC and HPLC. As the purification step progressed, it was confirmed that the antimicrobial activity was increased at the same concentration so it was judged purification. However, purification fold was different for each sensitive strains. It is estimated that the antimicrobial compounds produced by Lb. plantarum FC10 is not a single substance but at least two.
