본 연구는 여성의 손으로부터 분리한 두 유산균의 co-culture에 의한 향장활성에 대한 것이다. Lactobacillus rhamnosus (L. rhamnosus)와 Lactobacillus paracasei (L. paracasei)를 함께 배양하는 것은 최초이며, co-culture를 이용한 유산균은 tyrosinase 저해율이 L. paracasei와 L. rhamnosus에 비해 20.68%로 가장 높은 수치를 보였다. 또한, co-culture 유산균은 melanin 생성 저해율에서도 63.5%의 가장 높은 활성을 보였으며, 주름개선활성에 영향을 미치는 MMP-1의 생성량이 3726.3 pg/mL로 확인되었다. 반면 단일 유산균 배양시 얻어지는 MMP-1 생성량은 각각 L. rhamnosus가 13613.5 pg/mL, L. paracasei는 13012.0 pg/mL이 확인되었다. Collagen 생성량을 확인한 결과 co-culture 유산균이 380.7 ng/mL로 323.4 ng/mL의 양이 확인된 L. paracasei와 304.1 ng/mL의 양이 확인된 L. rhamnosus보다 많은 양이 생성됨을 확인하였다. 상기의 결과는 유산균의 co-culture가 향장활성 증진을 시킬 수 있다는 것을 나타내고, 이는 향장 활성에 영향을 미치는 항산화 활성을 측정한 결과 단일 유산균 배양을 통한 결과보다 co-culture 유산균의 항산화 활성이 더 높은 것으로 확인할 수 있었다.

In this study, cosmeceutical activities of the co-cultures of both lactic acid bacteria isolated from the hands of women, Lactobacillus rhamnosus (L. rhamnosus) and Lactobacillus paracasei (L. paracasei) were first reported: For whitening and anti-wrinkle activities, the co-culture showed the highest tyrosinase inhibition of 20.68%, compared L. paracasei and L. rhamnosus. The co-culture also showed the highest inhibition of melanin synthesis as 63.7%. In observing the anti-wrinkle activities of the co-culture, it generated only 3726.3 pg/mL of matrixmetalloproteinase-1 (MMP-1) production when 13613.5 pg/mL and 13012.0 pg/mL of MMP-1 production were estimated from L. rhamnosus and L. paracasei. Besides these, the extract from the co-culture yielded higher collagen production as 380.7 ng/mL, compared to 323.4 ng/mL and 304.1 ng/mL from L. paracasei and L. rhamnosus. These results indicate that the co-culture of both lactic acid could improve its cosmetic activities. This hypothesis was also confirmed that the co-culture of both bacteria showed strong antioxidant activities of DPPH free radical scavenging while the extract of L. rhamnosus and L. paracasei.

제안 방법
특히, 유산균 중에서 Lactobacillus균의 효능으로는 많은 연구가 되어 있으며 면역력 증진, 항균 효능, 피부미백 및 아토피성 피부염 개선효과 등이 보고된 바 있다[8,9]. 이에 착안하여 본 연구는 인체 피부에서 분리 및 동정하여 획득하게 된 균주 중에서 인체에 유익한 균으로 잘 알려진 유산균 Lactobacillus균 중 L. rhamnosus와 L.paracasei를 이용한 향장효능를 이용한 향장효능 평가를 실시하였으며, 그 효과를 증진시키기 위해 co-culture 방법을 적용하였다.
대상 데이터
실험에 쓰인 유산균 L. rhamnosus와 L. paracasei는 20 ~ 40대 남녀 40명의 손바닥을 MRS agar 배지에 찍어서 획득한 콜로니를 MRS broth에 옮겨 37 ℃에서 48 h을 배양하였다. 이때, 콜로니가 확연하게 나타난 20대 여성의 손바닥을 찍은 배지에서 배양한 균을 다시 MRS agar에 2차 접종을 실시하여, 획득한 콜로니를 16S rDNA sequence 분석을 통해 유산균 식별을 진행하여 신규 유산균 L.

하지만, 유산균들의 co-culture에 의해 균체수가 증가한다는 기존의 연구를 참고하면 co-culture를 통해 배양된 유산균은 그 균체량이 단일배양의 균체량 보다 많이 때문에 lipoteichoic acid을 비롯하여 세포막에 포함 된 인지질 등 그 외 활성 물질들에 의해 피부 미백 및 주름개선효능이 증가된 것으로 사료된다[15]. 다만, 기존에 연구되어진 유산균파쇄물에 대한 향장 활성의 경우 lipoteichoic acid나 substance P 등의 단리 물질의 연구 등 극히 일부만 연구가 되어 있기 때문에 향장 활성을 보이는 물질에 대한 추가 연구가 더 필요할 것으로 사료된다.
