The isolated strain exhibited superior resistance to heat, acid, and alkali compared with those of P. farinosa KCTC27412 as a control strain. The specific growth rate of P. farinosa MBY/L1569 at 46℃ was 0.37 ± 0.05 h−1, and the highest specific growth rate of 0.50 ± 0.02 h−1 was obtained when it was grown at pH 7.0 and 37℃ with 50 g/l (w/v) glucose as the carbon source. Under optimum growth conditions, strain MBY/L1569 produced ethanol 19.66 ± 0.68 g/l from glucose 50 g/l, with an approximate yield of 40%. P. farinosa MBY/L1569 was deposited at the Korean Collection for Type Cultures as pichia farinosa KCTC27753.

본 연구에서는 우리나라 전통 누룩으로부터 고온에서 성장속도가 빠르고 산, 알칼리에 대하여 내성이 우수한 효모를 선발하여 바이오에탄올을 생산하는데 적합한 균주를 확보하고자 하였다.
제안 방법
염색체의 Internal transcribed spacer 영역을 증폭하기 위하여 ITS1(5'-TCCGTAGGTGAACCTGCGC-3')와 ITS4(5'-TCCTCC GCTTATTGATATGC-3') 프라이머[7]를 사용하였다. PCR산물은 ㈜마크로젠(Korea)에 의뢰하여 염기서열을 해석하였으며, National Center for Biotechnology Information (NCBI, RP, MD, USA)의 BLAST (Basic Local Alignment Search Tool, http://www.ncbi.nlm.nih.gov/BLAST/) 프로그램을 사용하여 기존에 보고된 균주의 염기서열과 상동성을 비교하여 균주를 동정하였다.
누룩으로부터 분리된 P. farinosa MBY/L1569 균주의 생육 특성을 조사하기 위하여 배양온도, 배지의 초기 pH 및 탄소원으로 주입한 포도당 농도에 따른 비성장속도를 측정하였다. 모든 배양온도에서 대조구로 사용한 P.
1이 되도록 200 ml의 YEPD 배지에 접종하고 진탕배양기를 사용하여 37℃에서 200 rpm의 교반속도로 36시간 동안 배양하였다. 또한, 같은 조성의 YEPD 배지에 20 ml/l의 oxyrase (Bioworld, USA)를 첨가하고 플라스크를 밀봉하여 혐기적 배양조건 하에서 균주의 성장 및 포도당 소모 특성을 호기적 배양조건과 비교하였다.
분리된 균주의 생육특성을 조사하기 위하여 다양한 배양온도, pH 및 포도당 농도 조건에서 비성장속도를 측정하였다. 단일집락을 YEPD 배지 50 ml에 접종하여 30℃에서 18시간 동안 호기적인 조건으로 전배양하였다.
최적 배양온도를 결정한 후 최적 pH 조건을 설정하기 위하여 5 N H2SO4와 5 N NaOH를 사용하여 YEPD 배지의 초기pH를 조정한 뒤 배지에서 균주의 비성장속도를 측정하였다. 최적 배양온도 및 pH 조건을 결정한 후 탄소원으로 주입하는 포도당 농도를 조정하여 균주의 비성장속도를 측정하였다.
단일집락을 5 ml YEPD 배지에 접종하여 30℃에서 12시간 배양한 후, 세포흡광도(OD600) = 1에 해당하는 균체를 회수하였다. 회수한 균체는 멸균수로 5회 세척하고 100부터 104까지 단계적으로 멸균수를 사용하여 희석한 후 5 N H2SO4 (Korea) 용액과 5 N NaOH (Samchun, Korea) 용액을 사용하여 pH를 조정한 YEPD 평판배지에 15 μl씩 점적한 후 25, 30, 44℃에서 48시간 동안 배양하였다. 대조구로서 Pichia farinosa (Millerozyma farinosa) KCTC27412 균주를 한국 미생물자원센터(KCTC, Korea)로부터 분양 받아 사용하였다.

경상북도 상주시를 비롯하여 전국의 재래시장에서 판매되고 있는 누룩 12점을 구매하였다. 분쇄한 누룩 1 g을 멸균수 9 ml에 현탁한 후 101−105 희석배수까지 단계적으로 희석하고[2], chloramphenicol (100 mg/l, Sigma-Aldrich, USA)이 함유된 YEPD (yeast extract 10 g/l, peptone 20 g/l, glucose 20 g/l, pH 6.

모든 측정은 3회 반복하였으며 평균값과 표준오차는 SPSS (v 20.0, SPSS, USA)를 사용하여 결정하였고 유의성 검증은 Duncan의 다중범위 검정법을 사용하였다[24].
성능/효과
farinosa MBY/L1569 균주는 모든 pH 조건에서 성장하였지만 대조구 균주는 성장하지 못하여 P. farinosa MBY/L1569 균주가 대조구 균주보다 내열성과 내산성이 우수한 것으로 판단되었다. 배양온도 25℃와 30℃에서는 pH 5와 7로 조정한 배지에서 대조구와 실험구 균주가 유사하게 성장하여 유의적인 차이를 판단할 수 없었다.
평판배지를 이용한 실험결과로부터 P. farinosa MBY/L1569 균주가 대조구로 사용한 KCTC27412 균주에 비하여 내열성, 내산성이 우수한 것을 확인하였다.
P. farinosa MBY/L1569 균주는 37℃에서 가장 빠른 비성장속도(0.50 ± 0.02 h−1)를 나타냈다.
결론적으로 P. farinosa MBY/L1569 균주는 초기 pH 7.0, 포도당 농도 50 g/l, 배양온도 37℃에서 가장 빠른 비성장속도를 나타내었다.
farinosa KCTC27412 균주에 비하여 P. farinosa MBY/L1569 균주의 비성장속도가 유의적으로 우수하였다(Fig. 2A).
따라서 기존에 보고된 균주보다 고온에서도 우수한 성장을 보이며 산과 알칼리에 대한 내성이 우수한 P. farinosa MBY/L1569는 바이오에탄올 생산공정 등에 적용할 수 있는 가능성이 높고, 유전체 수준에서의 개량 등을 포함한 기반 연구에 사용할 수 있는 유망한 균주로 사료된다.
